NGF基因修饰的BMSCs对VaD大鼠海马区细胞凋亡及NMDAR1表达的影响

摘    要:目的探讨神经生长因子(NGF)基因修饰的骨髓间充质干细胞(BMSCs)对血管性痴呆(VaD)大鼠海马区细胞凋亡的影响。方法以NGF和(或)绿色荧光蛋白基因转染BMSCs;两血管法制备VaD模型。将大鼠随机分为假手术组、PBS组、BMSCs组及NCF修饰组。造模1周后,尾静脉注射NCF基因修饰的BMSCs;4周后行Morris水迷宫检测,观察其行为学改变;末端脱氧核苷酰基转移酶介导性dUTP切口末端标记法检测海马凋亡细胞,免疫组化法检测大鼠海马区神经细胞NGF、N-甲基-D-天门冬氨酸受体1(NMDAR1)表达。结果与PBS组、BMSCs组比较,NGF修饰组逃避潜伏期明显缩短,海马区神经元凋亡率明显下降,NMDAR1表达明显降低(P〈0.05或〈0.01)。结论NGF基因修饰的BMSCs对VaD大鼠的学习记忆能力有一定改善作用;对其海马细胞有一定保护作用;可降低VaD大鼠海马区NR1表达,提高NGF表达。
