安络化纤丸对小鼠肝纤维化治疗作用的实验研究

摘    要:目的观察中药安络化纤丸对四氯化碳(CCl4)小鼠肝纤维化的治疗作用,同时研究其可能的作用机制。方法选取64只雄性清洁级C57BL/6J小鼠,将其随机分为正常组、模型组、安络化纤组及理气活血组。给予正常组100%橄榄油剂1ml/kg腹腔注射,2次/周,共8周。其余3组分别给予20%CCl4橄榄油剂1ml/kg腹腔注射,2次/周,共8周。第5周起分别给予安络化纤组和理气活血组安络化纤丸和理气活血方灌胃0.2ml/10g,1次/d,共4周。同时给予正常组和模型组同等剂量的去离子水灌胃,1次/d,共4周。待实验完成时检测小鼠肝组织匀浆基质金属蛋白酶(MMP-1)及其抑制物(TIMP-1)浓度以及血清透明质酸(HA)、层黏连蛋白(LN)、三型前胶原(PCⅢ)、四型胶原(Ⅳ-C)水平,同时通过光镜观察小鼠肝脏病理组织学改变。结果安络化纤组、理气活血组肝组织匀浆TIMP-1浓度以及血清HA、LN、PCⅢ、Ⅳ-C水平均低于模型组,差异有统计学意义(P<0.05);安络化纤组肝组织匀浆TIMP-1浓度以及血清HA、LN、PCⅢ、Ⅳ-C水平均低于理气活血组,差异有统计学意义(P<0.05)。结论安络化纤丸对小鼠肝纤维化具有良好的治疗作用。其作用机制可能与降低肝组织匀浆TIMP-1浓度以及血清HA、LN、PCⅢ、Ⅳ-C水平有关。

Abstract:Objective To observe the therapeutic effect of traditional Chinese Medicine formula Anluohuaxianwan(ALHXW) on hepatic fibrosis in CCl 4 mice compared with traditional Chinese Medicine formula Liqihuoxue Fang(LQHXF),and to explore its mechanism.Methods Select 64 male clean grade C57BL/6J mice divide them randomly into 4 groups:the normal group,the model group,the ALHX group and the LQHX group.The normal group was abdominally injected with 100% olive oil of a dose of 1ml/kg twice a week for eight weeks.And the other three groups were abdominally injected with 20% CCl 4 olive oil of a dose of 1ml/kg twice a week for 8 weeks.Since the 5th week,the ALHX group and the LQHX group were intragastricly administered ALHXW Decoction and LQHX Decoction respectively with a dose of 0.2ml/10g once every day for 4 weeks.At the same time,the normal and the model groups were given the same volume of deionized water once a day for 4 weeks.After the end of experiment,detect the levels of liver tissue matrix metalloproteinases(MMP-1),its inhibitor(TIMP-1),serum hyaluronic acid(HA),laminin protein(LN),procollagen III(PC Ⅲ) and collagen IV(Ⅳ-C) by Enzyme-linked immunosorbent assay.And observe histological changes in mice liver pathology by light microscopy.Results The levels of TIMP-1,HA,LN,PCⅢ,Ⅳ-C of the ALHX and LQHX groups were both significantly lower than those of the model group(P 0.05).The levels of TIMP-1,HA,LN,PCⅢ,Ⅳ-C of the JPSG group were significantly lower than those of the HXHY group(P 0.05).Compared with the normal group,the MMP-1 levels of other three groups were all significantly higher(P 0.05).Conclusion ALHXW shows a striking effect of inhibition and treatment to hepatic fibrosis in mice caused by CCl 4.Its mechanism may be related to reducing the TIMP-1 levels,as well as serum HA,LN,PC Ⅲ,Ⅳ-C in the liver tissue homogenate.
