CD83+肿瘤浸润性树突状细胞在胃癌组织的表达及临床意义

目的肿瘤组织浸润树突状细胞(tumor infiltrating dendritic cells,TIDCs)数量和功能对患者预后有显著床意义,但CD83 TIDCs在胃癌组织中的表达情况尚不清楚,需要进行研究.方法采用免疫组化、原位杂交及流式细胞术检测45例胃癌TIDCs的CD83蛋白、CD83 mRNA以及S-100表达,并分析其临床意义.结果免疫组化显示胃癌组织(42/45)中有S100 散在表达;CD83主要表达于胃癌组织癌旁及正常胃黏膜上.原位杂交显示CD83 mRNA在少数标本中(7/45)表达.流式细胞术检测到低水平CD83表达(4%,45/45).S-100和CD83 mRNA表达数量与患者年龄、性别、肿瘤部位、浸润深度、病理分期及淋巴结转移情况无显著相关性.CD83蛋白免疫组化表达与胃癌TNM分期显著负相关(r=-0.924,P＜0.01);CD83流式细胞检查的表达率与胃癌TNM分期及淋巴结转移呈显著负相关(r=-0.879,P＜0.01;r=-0.782,P＜0.05).结论胃癌TIDCs CD83表达与胃癌TNM分期及淋巴结转移有关,可作为临床研究指标使用.

Expressions and clinical significance of CD83 + dendritic cells in gastric cancer tissue

Objective To detect the expression of S-100 and CD83 in gastric tumor tissues and evaluate their clinical significance as TIDCs markers in gastric cancer patients, since they can reflect the function of tumor infiltrating dendritic cells (TIDCs). Methods Immunohistochemistry (IHC), in situ hybridization (ISH) and flow cytometry were applied to detect the expression of S-100, CD83 mRNA and protein on DCs in 45 gastric adenocarcinoma tissues. The relationships of S-100 and CD83 expression with clinical pathological features was analyzed. Results IHC showed that S-100 expression was unevenly distributed within 42 cases of cancer tissues, but CD83 only expressed in para-cancerous tissues and normal tissues. ISH showed that CD83 mRNA was limitedly expressed in 7 samples. S-100 expression had no significant correlation with clinical pathological features while CD83 correlated with TNM stages reversely. Detected by flow cytometry, CD83 was low level expressed in all 45 gastric cancer tissues and negative correlations were found with lymphoid metastasis and TNM stages of gastric cancer(r=-0.879, P<0.01; r=-0.782, P<0.05). Conclusion TIDCs CD83 expression correlated with lymphoid metastasis and TNM stages of gastric cancer, and could be used as a marker in further research.
