摘    要:
背景:研究显示α-玉米赤霉醇对绝经后骨质疏松症具有明显的防治作用。目的:观察α-玉米赤霉醇对体外大鼠成骨细胞骨保护素分泌及其mRNA表达的影响。方法:将体外培养的成骨细胞中分别加入雌激素和α-玉米赤霉醇(10-5~10-10 mol/L)进行干预。结果与结论:ELISA法和RT-PCR检测发现,不同浓度的α-玉米赤霉醇(10-5~10-10 mol/L)作用于成骨细胞后,骨保护素的分泌量及细胞骨保护素mRNA表达较对照组均增加(P < 0.05)。结果证实,在骨髓基质细胞来源的成骨细胞中加入α-玉米赤霉醇可促进成骨细胞骨保护素的分泌及其mRNA的表达。

Abstract:
BACKGROUND: Studies showed that α-zeranol (α-ZAL) has significant prevention and treatment effect on postmenopausal osteoporosis.OBJECTIVE: To observe the effects of α-ZAL on the secretion of osteoprotegerin (OPG) and expression of OPG mRNA in osteoblasts of the rats. METHODS: Estrogen and α-ZAL (10-5-10-10 mol/L) were added into in vitro cultured osteoblasts respectively for intervention.RESULTS AND CONCLUSION: The results of ELISA and RT-PCR showed that after application of different concentrations of α-ZAL (10-5-10-10 mol/L), the secretion of OPG and expression of OPG mRNA in the estrogen group were higher than those in the control group (P < 0.05). The results confirmed that in the bone marrow mesenchymal stem cells-derived osteoblast, adding α-ZAL can promote osteoblast OPG secretion and mRNA expression.
