| 摘 要： | 目的 分析胃癌组织中miRNA-191的表达及预测其靶基因.方法选取胃癌及配对癌旁正常组织标本50例,应用基于SYBR GreenⅠ的实时荧光定量PCR(qRT-PCR)检测miRNA-191的表达情况,后用2-ΔΔCT方法进行相对定量分析.利用生物信息学软件对miRNA-191进行靶基因预测.结果 miRNA-191在胃癌组织中的表达显著高于癌旁正常组织(P＜0.05),miRNA-191与临床病理参数(性别、年龄、病理分型、淋巴结转移和TNM分期)无明显关系(P＞0.05).对miRNA-191预测得到PLCD1、SOX4等9个靶基因.结论 miRNA-191在胃癌组织中高表达,可能在胃癌的发病中起重要作用.

| Abstract: | Objective To investigate the expression of miRNA-191 in gastric carcinoma tissues.Methods The expression of miRNA-191 in both gastric carcinoma and their adjacent non-tumour tissues was quantified with SYBR Green Ⅰ based qRT-PCR according to the formula of 2-ΔΔCT.The protential target genes of miRNA-191 were predicted by using MiRanda,TargetScan and PicTar.Results The expression of miRNA-191 was significantly up-regulated in gastric carcinoma tissues than their adjacent non-tumour tissues(P<0.05).There was no significant correlation between miRNA-191 expression and clinicopathologic parameters,including age,gender,histological type,lymph node metastasis and clinical stage.Nine predicted target genes,including NDST1 and SOX4,had been confirmed as direct targets of miRNA-191.Conclusion Over-expression of miRNA-191 in gastric carcinoma is correlated with pathogenesis of gastric carcinoma. |
