乳腺癌组织HER-2扩增状态及其与组织微血管生成拟态的相关性

摘    要:
目的 探讨乳腺癌人类表皮生长因子受体2 (HER-2)蛋白表达及基因扩增状态与组织微血管生成拟态(VM)的关系.方法 选取已确诊乳腺癌患者的石蜡标本组织200例,观察肿瘤组织中VM病理改变.采用免疫组化和荧光原位杂交法检测肿瘤组织中HER-2蛋白和基因的扩增情况,采用CD34和PAS双重染色法观察肿瘤组织中VM的情况,分析HER-2蛋白表达和基因扩增状态与VM的关系,分析HER-2蛋白表达、基因扩增及VM与乳腺癌临床病理参数的相关性.结果 乳腺癌组织中HER-2蛋白阳性率和基因扩增率分别为67.5%(135/200)、19.0% (38/200).HER-2蛋白表达及HER-2基因扩增与肿瘤大小和组织学分级相关(P均<0.05),VM阳性与淋巴结转移和组织学分级相关(P均<0.05).结论 乳腺癌组织中HER-2的表达与肿瘤的恶性进展存在相关性,且HER-2的表达可以促进乳腺癌组织中VM的形成,VM的存在与组织学分级和淋巴结转移关系密切.

Abstract:
Objective To investigate the expression of human epidermal growth factor receptor 2 (HER-2) protein and gene amplification,and its relationship with vasculogenic mimicry (VM) in breast cancer.Methods A total of 200 cases of patients with breast cancer were selected,and their VM pathological changes were observed.The expression of HER-2 protein and gene amplification was detected by immunohistochemisrty and fluorescence in situ hybridization.The expression of VM was detected with perform double staining of PAS and CD34.The relationship of HER-2 and gene amplification with VM was analyzed as well as the correlation between the expression of HER-2 protein,gene amplification and clinic pathological parameters of breast cancer,Results The positive rate and amplification rate of HER-2 gene in breast carcinoma were 67.5% (135/200) and 19.0% (38/200),respectively.Both of them were associated with tumor size and histological grades (all P < 0.05).Positive VM was also associated with lymph node metastasis and histological grades (all P < 0.05).Conclusion HER-2 expression in breast cancer tissues is associated with tumor malignant progress,and it can promote the formation of the microvascular mimicry.The expression of VM is also closely related to histological grades and lymph node metastasis.
