解毒化浊方诱导HER-2阳性乳腺癌SK-BR-3细胞凋亡的研究

摘    要:目的采用解毒化浊方血清干预乳腺癌SK-BR-3细胞,观察中药血清诱导乳腺癌细胞凋亡的作用。方法 8只雌性Wistar大鼠分为对照组、中药组、西药组、联合组,每组2只,给药制备含药血清,中药血清分24h、48h时间干预SK-BR-3细胞,采用流式细胞技术检测含药血清干预乳腺癌细胞后细胞凋亡率。结果含药血清干预SK-BR-3细胞24h后,与对照组比较,各用药组早期细胞凋亡率、总凋亡率明显增加,西药组(4.76±0.63)%、(7.00±1.75)%]、联合组(6.93±0.76)%、(9.93±0.28)%]较对照组(1.03±0.23)%、(2.53±0.42)%]增加明显(P0.05);晚期细胞凋亡率各组差异不明显(P0.05)。干预SK-BR-3细胞48h后,各用药组早期细胞凋亡率(4.76±0.23)%、(8.56±0.64)%、(6.80±0.10)%]与对照组(1.60±0.10)%]比较,差异有统计学意义(P0.05);晚期细胞凋亡率、总凋亡率比较,西药组(32.90±2.00)%、(41.46±1.93)%]、联合组(22.86±9.74)%、(34.83±6.51)%]高于对照组(1.93±0.20)%、(3.53±0.13)%](P0.05),西药组与联合组细胞凋亡率、总凋亡率比较,差异无统计学意义(P0.05)。结论解毒化浊方可能有诱导乳腺癌SK-BR-3细胞凋亡,协同抑制剂减毒增效作用。

Experimental Study on Jiedu Huazhuo decoction Induction of Apoptosis in HER-2 Positive Breast Cancer SK-BR-3 Cells
Abstract:Objective: To observe the effect of Jiedu Huazhuo decoction on human breast cancer cells SK-BR-3 and to observe the effect of traditional Chinese medicine in inducing apoptosis of breast cancer cells and to explore the effect of detoxification turbidity on HER-2 overexpression breast cancer Mechanism. Methods: Female Wistar rats were divided into 4 groups, which were respectively administrated with drug-containing serum. Serum was administered to SK-BR-3 cells for 24 h and 48 h respectively. Flow cytometry was used to detect the apoptosis rate of Chinese herbal serum after intervention of breast cancer cells. Results: Compared with the control group, the apoptosis rate and the total apoptosis rate of SK-BR-3 cells were significantly increased in the drug-treated groups compared with the control group, especially in the Western medicine group and the combined group (P < 0.05). There was no significant difference between late apoptosis rate in each group (P> 0.05). Compared with the control group, the early apoptosis rate of SK-BR-3 cells was significantly different (P < 0.05) after intervention with SK-BR-3 cells for 48h. The apoptosis rate and the total apoptosis rate of the latter groups were significantly higher than those of the western medicine group and the combined group The control group was especially obvious, with significant difference (P<0.05). Conclusion: Jiedu Huazhuo decoction can induce the apoptosis of SK-BR-3 breast cancer cells and synergize with the attenuator and synergistic effect of the inhibitor to exert antitumor effect.
