醋酸铅对小鼠胸腺细胞凋亡及Bax和Bcl-2基因表达的影响

目的探讨醋酸铅诱导小鼠胸腺细胞凋亡及对Bax和Bc l-2基因表达的影响。方法以3种不同浓度的醋酸铅灌胃饲养小鼠46 d后,取胸腺组织,用琼脂糖凝胶电泳法检测胸腺细胞凋亡情况,Western blot法检测胸腺组织中Bax和Bc l-2的表达情况。结果铅处理组小鼠胸腺细胞有被降解的凋亡带,并且降解条带的量随着染铅剂量的增加而增加;小鼠胸腺细胞Bax表达量均高于对照组,并且表达量随着染铅剂量增加而升高(P<0.05);而Bc l-2表达量均低于对照组,且表达量随着染铅剂量的增加而递减(P<0.05),Bc l-2/Bax表达比明显降低(P<0.01)。结论醋酸铅可诱发小鼠胸腺细胞凋亡,,促进胸腺细胞中Bax基因的表达,抑制Bc l-2基因的表达,降低Bc l-2/Bax表达比。

Effects of plumbi acetas on apoptosis and gene expression of Bax and Bcl-2 of thymus cell in mice

Objective To investigate effects of plumbi acetas on apoptosis and gene expression of thymus cell in mice.Methods Mice were performed with intragastric administration 3 kinds of concentration of plumbi acetas for 46 days,thymus tissues were obtained.Apoptosis of thymus cell was detected by agarose gel electrophoresis and expression of Bax and Bcl-2 in thymus by Westen blot.Results Degradated apoptosis band in thymus cell of the group treated with plumbi acetas,the amount of degradated band was increased with increase of plumbi acetas.Expression of Bax in observation group was higher than that in control group and amount of expression was increased with the plumbi acetas(P<0.05).Expression of Bcl-2 in observation group was lower than that in control group and amount of expression was decremented with plumbi acetas(P<0.05).Ratio of Bcl-2 versus Bax obviously decreased(P<0.01).Conclusion Pumbi acetas probably induces the apoptosis of the thymus cell in mice,promotes the Bax expression and inhib Bcl-2 expression of the thymus cell and reduces the ratio of Bcl-2/Bax.
