摘    要:目的比较不同压力CO2气腹处理的人胃癌MKN-45细胞在裸鼠腹腔内的成瘤数量、重量及裸鼠生存时间,探讨CO2气腹对胃癌细胞侵袭转移能力的影响。方法实验分为4组(对照组和5、10、15mmHg CO2气腹组),即将胃癌细胞在5%CO2 37℃条件下或5、10、15mmHg100%CO2 37℃条件下培养4h,然后收集细胞并将2×10^5个细胞注射入裸鼠腹腔,建立胃癌腹腔转移动物模型。4周后处死裸鼠,观察其腹腔成瘤率、成瘤数量和重量以及裸鼠生存时间。结果各组裸鼠腹腔成瘤数量、肿瘤总重量和生存时间均无显著差异(P〉0.05)。结论在气腹压力≤15mmHg时CO2对胃癌MKN-45细胞在裸鼠腹腔内的种植转移无显著影响。

Effects of CO2 Pneumoperitoneum on the Intra Abdominal Implantation of Gastric Cancer Cells in Animals

Abstract:Objective To compare the effects of CO2 insufflation administered at different pressures on the intra-abdominal implantation of cultured human gastric cancer cells and survival time in nude mice, and investigate the influence of CO2 pneumoperitoneum on the invasive ability of the cancer cells. Methods Sixty-four nude mice were equally divided into 4 groups: 5, 10, and 15 mm Hg CO2 insufflation groups and a control group. MKN-45 gastric cancer cells were culture for 4 hours at 37 ℃ with 5% or 5, 10, or 15 mm Hg 100% CO2. Afterwards, nude mice models of intra-abdominal implantation of gastric cells were established by injecting 2 × 10^6 cells that cultured at different conditions to different groups. Four weeks later, the mice were killed to explore the rate of tumor formation, and number and weight of the intra-abdominal cancer nodules. The survival time in each group was compared as well. Results No statistically significant difference was detected in the number of implanted tumors, weight of the nodules, and survival time of the nude mice among the four groups ( P 〉 0. 05 ). Conclusion CO2 pneumoperitoneum with a insufflation pressure≤ 15 mm Hg do not increase the implantation of MKN-45 gastric cancer cells in the abdominal cavity of nude mice.
