摘 要: | 目的 观察以壳聚糖一胶原偶联的生物膜为载体,联合移植雪旺细胞(SCs)与神经干细胞(NSCs)在坐骨神经损伤修复过程中所起的作用,评价该方法的疗效.方法 分离纯化SCs和NSCs,传代4代后共培养于壳聚糖一胶原生物膜上.32只Wistar成鼠(体重350~400 g)建立坐骨神经缺损动物模型,随机分为4组:空白组;SCs移植组;NSCs移植组;NSCs联合SCs移植组.14周后观察Cy3荧光素染色、MBP免疫酶染色及苏木素-伊红(HE)染色的结果.结果 BDA示踪显示术后联合移植组和SCs组有较多的神经纤维穿过缺损部位,大鼠右后肢感觉和运动功能恢复较其他组明显,修复神经段神经传导速度比值也大于其他组(P<0.05).SCs组和联合移植组间差异无统计学意义(P>0.05).结论 NSCs和SCs与壳聚糖一胶原生物膜相容性良好,壳聚糖一胶原生物膜介导的SCs联合NSCs桥接周围神经缺损可使部分神经再生.

Connecting the defects of sciatic nerve by transplantation of Schwann cells and neural stem cells embedded to the chitosan-collagen complex film |

Abstract: | Objective To evaluate the effect of bridging the defects of sciatic nerve in Wistar rats by transplantation of Schwann cells (SCs) and neural stem cells (NSCs) embedded to the chitosan-collagen complex film. Methods SCs and NSCs were isolated and purified. These clones were passaged continuously for 4 generations. Then the two types of cells were embedded to the chitosan-collagen complex film. Thirty-two adult Wistar rats, weighing 350-400 g, were divided into 4 groups at random:Blank group, SCs group,NSCs group and SCs + NSCs group. An about 10 mm gap of right sciatic nerve was cut in all rats. The films planted by SCs and/or NSCs or none were transplanted to bridge the sciatic nerve defects. Fourteen weeks later MBP was detected by using cy3 fluorescence staining, HE staining, and immunohistochemistry staining. The velocity of electrical conduction of sciatic nerves was calculated. Results The recovery of sense and motion function in SCs + NSCs group and SCs group were better than the other two groups. SCs + NSCs group and SCs group were superior to the other two groups on neurofibril regeneration and velocity of electrical conduction of sciatic nerve ( P > 0.05 ). But there was no significant difference between SCs group and SCs + NSCs group. Conclusion The two types of cells have a good compatibility with the chitosan-collagen complex film. Transplantation of SCs and NSCs embedded to the chitosan-collagen complex film can guide the regeneration of neurofibrils. |
