Beclin-1在肺腺癌发生发展中的作用及其对预后的影响

目的研究自噬特异性标志物Beclin-1在肺腺癌发生、发展中的变化及其对术后患者预后的影响。方法采用反转录-聚合酶链反应(RT-PCR)及Western blot法分别检测108例正常肺组织、肺不典型腺瘤样增生(AAH)、肺原位腺癌、肺微浸润性腺癌及浸润性腺癌组织标本中自噬特异性标志物Beclin-1在基因及蛋白水平的表达,比较Beclin-1在不同组织中及不同病理分期肺腺癌患者肿瘤组织中的变化,分析其对肺腺癌患者术后预后的影响。结果肺腺癌Beclin-1基因及蛋白水平的表达均显著低于正常肺组织(P0.05),浸润性肺腺癌组织表达最低;早期肺腺癌(Ⅰ~Ⅱ期)患者肿瘤组织Beclin-1的表达高于中晚期肺腺癌(Ⅲ~Ⅳ期)患者(P0.05)。肺腺癌伴淋巴结转移者肿瘤组织Beclin-1的表达低于淋巴结阴性患者(P0.05)。肺腺癌肿瘤组织Beclin-1阳性表达患者术后生存时间显著高于Beclin-1阴性表达者(P0.05)。结论 Beclin-1的表达与肺腺癌的发生、发展相关;肺腺癌Beclin-1的表达缺失预示肺腺癌患者术后生存率较差。Beclin-1可能成为预测肺腺癌患者术后预后的重要标志物。

The role of Beclin1 in the occurrence and development of lung adenocarcinoma and its effect onprognosis

Objective To study the changes of Beclin1 in the occurrence and development of lung adenocarcinoma and its effect on prognosis. Methods The expressions of autophagy specific marker Beclin1 in gene and protein levels were detected by RT-PCR and Western blotting. Normal lung tissues,a-typical hyperplasia(AAH),lung adenocarcinoma in situ,micro-invasive adenocarcinoma lung cancer and invasive adenocarcinoma lung cancer were detected respectively. The changes of Beclin1 in different tis-sues and pathological stages were compared to evaluate their effects on prognosis. Results The expression of Beclin1 in lung adenocarcinoma was significantly lower than that in the normal lung tissues(P < 0. 05) and expression in the invasive adenocarcinoma lung cancer tissues was the lowest. The expression of Bec-lin1 in tumor tissues of patients with early stage tumors(stage Ⅰ ~ Ⅱ)was significantly higher than that in patients with advanced tumors(stage Ⅲ ~ Ⅳ stage)(P < 0. 01). The expression of Beclin1 in lung adeno-carcinoma with lymph node metastasis was lower than that in lymph node negative patients(P < 0. 05). The survival time of patient with positive Beclin1 expression was significantly higher than that of patient with negative expression(P < 0. 05). Conclusion The expression of Beclin1 was related to the occur-rence and development of lung adenocarcinoma. The absence of Beclin1 in lung adenocarcinoma indicates that the survival rate of patients is poor. Beclin1 may be an important marker for predicting the prognosis of patients with lung adenocarcinoma.
