摘    要:IgE类型的抗体是过敏性疾病的始动因素,抗IgE抗体能特异性地识别并结合游离IgE分子,下调IgE分子水平,人源化抗IsE单克隆抗体的出现更是为过敏性疾病带来了新的治疗方法。临床研究治疗哮喘、过敏性鼻炎以及食物过敏症等疾病中获得的大量数据更显示了其良好的疗效。与传统临床治疗手段相比,人源化抗IgE抗体具有副作用小,安全性高等优点。此外,治疗过敏性疾病的拮抗分子的研究也十分盛行。在分析IgE分子功能表位的基础上进行适当的分子设计可获得潜在的治疗过敏性疾病的拮抗剂如抗IgE抗体类似多肽、IgE类似肽以及其他抑制性小分子等。

Abstract:Immunoglobulin E(IgE),the antibody responsible for allergic hypersensitivity,can be identified and down-regulated by anti-IgE antibodies.Monoclonal humanized anti-IgE has put forward a fundamentally new concept for the therapy of allergic diseases.Recent data from clinical studies with anti-IgE have documented its efficacy in the treatment of asthma,allergic rhinitis and food allergy.Compared with traditional clinical treatments,anti-IgE monoclonal antibodies excel at its low side-effects and high safety.Additionally,antagonistic peptides,which inhibit crucial molecules of allergic hypersensitivity,are involved in the structure-based therapy studies.Molecular design based on the functional epitope analysis of IgE could obtain potential antagonists of allergic diseases such as peptides derived from anti-IgE antibodies,IgE structural analogous peptides and suchlike molecules.
