肺癌中mPGES-1的表达及生物学意义

摘    要:目的 探讨前列腺素E合成酶(mPGEs-1)在肺癌中的表达及生物学意义,为肺癌发生和发展的分子机制提供理论依据.方法 采用RT-PCR检测肺癌及癌旁组织中mPGEs-1mRNA的表达,同时采用免疫组织化学S-P法检测相应标本中mPGEs-1蛋白的表达.结果 肺癌组织中mPGEs-1 mRNA的表达量明显高于癌旁组织(P<0.05).mPGEs-1蛋白阳性表达率肺癌组织中为66.7%(40/60),在相应癌旁组织中为10%(6/60),肺癌明显高于癌旁(x2=20.38,P<0.01).肺癌mPGES-1蛋白的阳性表达率与肿瘤大小、组织类型无关(P>0.05);而与分化程度、淋巴结转移、TNM分期有关,随着组织分化程度的降低、淋巴结的转移和临床分期的增加,肺癌mPGEs-1蛋白的阳性表达率逐渐增高,差异有显著性(P<0.05).结论 mPGEs-1的高表达可能在肺癌的发生发展中起着重要作用,它们可能为肺癌的早期诊断和为开发肺癌的靶向治疗提供一定的临床依据.

Abstract:Objective To explore the expression and biological significance of mPGEs - 1 in lung cancer and to provide a theoretical basis for the molecular mechanism of the development of lung cancer. Methods The expressions of mPGEs - 1 in mRNA level in cancerous tissue and tumor - adjacent tissue were detected by RT - PCR, while the immunohistochemical S - P method was used to evaluate the expression of mPGEs - 1 proteins in the corresponding tissues. Results The mRNA level of mPGEs - 1 significantly increased(P〈 0.05) in lung cancer group as compared with those in the tumor - adjacent tissue. The positive expression rate of mPGEs - 1 protein in cancerous tissue and tumor 〉 adjacent tissue were 66.7% (40/60)and 10% (6/60), respectively. The positive expression rate of mPGEs - 1 was significantly higher in lung cancer than in tumor - adjacent tissue ( P 〈 0.001). The positive expression rate of mPGEs - 1 protein in lung cancer was independent of size of tumor and histological type(P〉0. 05), but correlated with histological grade, lymph node metastasis and TNM staging. The positive expression rate of mPGEs - 1 protein increased gradually with lymph node metastasis, the decrease of histological grade and the increase of TNM staging. There was a significant difference(P〈 0.05). Conclusion It suggested that the expression of mPGEs - 1 in lung cancer may play an important role in the process of carcinogenesis, and may provide a clinical basis for the early diagnosis and targeted therapy of lung cancer.
