uPAR表达对鼻咽癌侵袭和转移的影响

目的　探讨尿激酶型纤溶酶原激活剂受体 (uPAR)对鼻咽癌侵袭和转移的影响。方法　以鼻咽癌细胞系 (CNE 2Z)及其克隆株 (CNE 2Z H 5 ,CNE 2Z H 5 9)作为研究材料 ,应用逆转录聚合酶反应 (RT PCR )检测uPAR在基因转录的表达 ;应用蛋白免疫印迹 (Westen Blotting)方法检测uPAR蛋白水平的表达 ;通过异质黏附抑制实验 ,观察uPAR在鼻咽癌细胞侵袭和转移过程中的作用。结果　uPAR在CNE 2Z H 5 9中表达最高 ,其次为CNE 2Z H 5 ,在CNE 2Z中表达最低 ;uPAR抗体能抑制CNE 2Z H 5 9的异质黏附能力。结论　uPAR可能促进鼻咽癌细胞的侵袭和转移

Effect of uPAR on Invasion and Metastasis of Nasopharynal Carcinoma(NPC)

Objective To explore the effect of uPAR on invasion and metastasis of NPC.Methods A Nasopharyneal carcinoma cell-line(CNE-2Z) and its sub-line(CNE-2Z-H5,CNE-2Z-H5-9) with different ability of lymph node metastasis were used.The expression of uPAR was detected by RT-PCR at level of Mrna and by Western-Blotting at protein level.The effect of uPAR on invasion and metastasis of PNC was observed through the experiment of heterogeneous adhesion.Results mRNA expression of uPAR was the highest in CNE-2Z-H5-9,then was in CNE-2Z-H5 whereas the lowest in CNE-2Z.uPAR antibody inhibited the heterogeneous adhesion of CNE-2Z-H5-9.Conclusion uPAR may promote the metastasis and invasion of NPC.
