摘    要:目的:建立测定人血浆中万古霉素浓度的高效液相色谱法。方法:采用外标法定量,万古霉素血浆样品经5%高氯酸处理,采用Inertsil ODS-SP C18(150 mm×4.6 mm,5μm)色谱柱,流动相为乙腈-0.025 mol/L磷酸二氢钾缓冲液(pH 3.2)(11.5∶88.5),流速为0.5 ml/min,检测波长为230 nm,柱温30℃,进样量20μl。结果:血浆中万古霉素在1.56~100.00 mg/L内线性良好(r=1),检测限0.33 mg/L(S/N=3),平均回收率为95.26%,RSD为1.16%。结论:该方法灵敏、准确、快速、操作简便、取样量少,适合临床万古霉素血药浓度的监测。

Abstract:Objective: To establish a method of determining vancomycin in human plasma by HPLC. Methods: Using the external standard method,the chromatography was performed on Inertsil ODS- SP C18 column( 150 mm × 4. 6 mm,5 μm) with the mobile phase consisting of acetonitrile- 0. 025 mol / L potassium dihydrogen phosphate buffer solution( 11. 5∶88. 5,p H 3. 2),the flow rate was 0. 5 ml / min,the detection wavelength was at 230 nm,the column temperature was 30 ℃,and the injection volume was 20 μl. Results: In the plasma,the calibration curve was linear within the range of 1. 56 ~ 100. 00 mg / L( r = 1). The limit of test was 0. 33 mg / L( S / N = 3),The average relative recovery of low concentration,moderate concentration and high concentration was 95. 26%,and average intra- day and inter- day RSD were 1. 31 and 1. 66%. Conclusion: The method developed in this study is rapid,accurate and sensitive,it is suitable for the determination of vancomycin in human plasma in clinical practice.
