曲古菌素A联合5-氮基-2-脱氧胞苷对人胃癌SGC-7901细胞生长及其TESTIN基因表达的影响

摘    要:目的探讨曲古菌素A(TSA)联合5-氮基-2-脱氧胞苷(5-aza-dC)对人胃癌细胞(SGC-7901)生长及其TESTIN(TES)基因表达的影响。方法将实验分为空白对照组(不加任何药物处理)、TSA(150ng/ml)、5-aza-dC(5μmo1/L)、TSA(150ng/ml)+5-aza-dC(5μmo1/L)共4组,用MTT法测定各组药物作用对SGC-7901细胞生长的影响,用流式细胞仪分析细胞周期,AnnexinV/PI检测细胞凋亡,RT-PCR分析SGC-7901细胞TESmRNA表达。结果曲古菌素A和(或)5-aza-dC分别作用24h、48h、72h后均能抑制细胞生长,联合组较单药组抑制率显著增强,且随着药物作用时间的延长,抑制率增强更明显;流式细胞仪分析及AnnexinV/PI凋亡检测发现干预胃癌细胞48h后,联合组G0/G1期比率为(85.23±2.17)%,较单药组显著增多(P〈0.05);同时诱导细胞凋亡,联合组细胞凋亡率达到(32±3.25)%,比单药组明显增加(P〈0.05);TSA和(或)5-aza-dC干预胃癌细胞48h后,联合组较单药组显著增强TESmRNA的表达(P〈0.05)。结论曲古菌素A联合5-aza-dC干预可抑制SGC-7901细胞生长,阻止细胞周期于G0/G1期并促进细胞凋亡,抑癌基因TESmRNA表达增多均较单药组明显。

Abstract:Objective To study the combination of 5-aza-2-deoxycytidine and trichostatin on the Expression of TESTIN(TES) gene and the growth of human gastric cancer cells.Methods Experimental cells were divided into four groups by received different drugs:the control group without using any drug,group with 150ng/ml trichostatin,the group with 5 μmol/L 5-aza-2-deoxycytidine and the group with 150ng/ml trichostatin and 5 μmol/L 5-aza-2-deoxycytidine in combination.Cell growth was evaluated by MTT and cell cycle by flow cytometry,apoptosis by AnnexinV/PI fluos stainging and RT-PCR was applied to detect the TESmRNA.Results Trichostatin and (or) 5-aza-2’-deoxycytidine all inhibited the growth of SGC-7901 cells after being treated for 24,48,72 h.The inhibitory rate in the union medication group was significantly higher than that in the independent medication group.The inhibitory rate was increased in a time-dependent manner.Flow cytometry and AnnexinV/PI apoptosis detection:After SGC-7901 cells were treated with TSA and (or) 5-aza-dC for 48h,the rate of G0/G1 phase in the union medication group was (85.23±2.17)%,which was significantly higher than that in the independent medication group.Besides,the apoptosis rate in the union medication group was about (32±3.25)%,which was higher than that in the independent medication group.The TESmRNA expression in SGC-7901 cells was much more in the union medication group than that in the independent medication group in 48 h.Conclusion In the union medication group the drug was significantly much more effective than that in the independent medication group in inhibiting the proliferation of SGC-7901 cells,arresting cells in G0/G1 phase,inducing cells apoptosis and up-regulating TESTINmRNA expression.
