冬凌草甲素对人乳腺癌Bcap-37细胞的增殖抑制作用及其机制

摘    要:目的观察冬凌草甲素对人乳腺癌细胞株Bcap-37细胞的生长抑制作用,初步探讨冬凌草甲素调控乳腺癌细胞增殖的分子机制。方法 MTT比色法检测冬凌草甲素对Bcap-37细胞的增殖抑制作用,相差显微镜观察细胞形态变化,流式细胞仪分析细胞周期分布,RT-PCR检测p21、cdc2和cyclin B1 mRNA表达。结果冬凌草甲素对Bcap-37细胞有显著的增殖抑制作用,呈时间剂量依赖趋势(P〈0.01);处理后的Bcap-37细胞发生明显的形态改变;作用24h后,Bcap-37细胞在G2/M期阻滞;冬凌草甲素作用Bcap-37细胞后,p21 mRNA水平的表达增加,而cdc2、cyclin B1 mRNA的表达降低(均P〈0.05)。结论冬凌草甲素可抑制乳腺癌细胞的增殖,该作用可能与p21 mRNA表达上调及cdc2、cyclin B1 mRNA表达下调有关。

Abstract:Objective To investigate the inhibitory effect of oridonin on human breast cancer cell line Bcap-37 and its mechanisms.Methods Growth inhibition rate of Bcap-37 cells was measured with MTT method.Morphological changes in carcinoma cells were defected by light microscope.The cell cycle were determined by flow cytometry.The mRNA expressions of p21,cdc2 and cyclinB1 were assayed by RT-PCR.Results Oridonin significantly inhibited the proliferation of Bcap-37 human breast cancer cells in a time-dose dependent manner.After dealed with oridonin,the morphology of Bcap-37 cells changed.Bcap-37 cells were arrested at G2/M phase.RT-PCR indicated that the mRNA expressions of p21 was increased,while the levels of cdc2 and cyclinB1 mRNA expressions were decreased(P0.05).Conclusion Oridonin can inhibit proliferation of breast cancer cells and the mechanisms possiblely is associated with down-regulating the mRNA expressions of p21 and up-regulating the mRNA expressions of cdc2 and cyclinB1.
