胶质瘤患者肿瘤组织中IGF2BP2的表达及临床意义

目的探讨胰岛素样生长因子2结合蛋白2（insulin-like growth factor2 binding protein2 IGF2BP2）在脑胶质瘤中的表达状况。方法应用实时荧光定量PCR技术检测47例不同病理级别（9例Ⅱ级,21例Ⅲ级,17例Ⅳ级）胶质瘤组织IGF2BP2表达情况,同时采用免疫组化法检测47例肿瘤组织中IGF2BP2的表达情况。结果Ⅱ级胶质瘤IGF2BP2蛋白阳性表达率阳性率（100%）明显高于胶质瘤Ⅲ级（52.38%）（P=0.013）和Ⅳ级（11.76%）（P=0.000）胶质瘤Ⅲ级高于Ⅳ级（P=0.015）;Ⅱ级胶质瘤IGF2BP2的基因表达量（0.075±0.020）高于Ⅲ级（0.047±0.011）（P=0.016）和Ⅳ级胶质瘤（0.026±0.004）（P=0.000）,Ⅲ级胶质瘤IGF2BP2的基因表达量高于Ⅳ级胶质瘤（P=0.000）。结论在Ⅱ级胶质瘤组织中IGF2BP2的表达水平高,并且随着病理级别的增高表达减弱,推测IGF2BP2可能在胶质瘤的发生发展中扮演重要的角色。

The Expression and Clinical Significance of IGF2BP2 Gene in Tumor Tissues of Patients with Glioma

Objective To in tumor tissues of patients explore the expression of insulin - like growth factor2 binding protein2 （ IGF2BP2 ） with glioma. Methods The expression of IGF2BP2 gene of different pathological grade glioma tissue （9 cases of Grade Ⅱ,21 cases of Grade III,and 17 cases of Grade IV） were detected using real - time PCR technology while the expression of IGF2BP2 gene was detected using immunohistochemistry. Results Positive rate of IGF2BP2 protein expression in different levels of gliomas as followed ： positive rate of level Ⅱ was 100%, positive rate of level III was 52.38% and positive rate of level IV was only 11.76%. Posi- tive rate in IGF2BP2 protein expression of grade II was significantly higher than that of grade III and grade IV （ P 〈 0.05 ） while positive rate in IGF2BP2 protein expression of grade III was significantly higher than that of grade IV （P 〈 0.05 ）. IGF2BP2 gene expression volume in different levels of gliomas as followed ：the expres- sion volume of level Ⅱ was （0. 075 ±0. 020） ,the expression volume of level Ⅲ was （0.047 ±0. 011 ） and the expression volume of level 1V was （0.026 ± 0. 004）. Conclusion The IGF2BP2 expression may play an im- portant role in glioma development process and it can be used for further study.
