外阴鳞癌中Ki67和Caspase-3的表达及相关性

目的:探讨Ki67和Caspase-3在外阴鳞状上皮细胞癌中的表达、临床意义及相关性。方法:选取外阴鳞癌(VSCC)30例,正常外阴皮肤10例,收集外阴鳞癌患者的临床病理资料进行统计分析。采用鼠抗人Ki67单克隆抗体及兔抗人Caspase-3多克隆抗体用S-P免疫组化法研究ki67及Caspase-3。结果:Ki67在外阴鳞癌中表达阳性率为83.3%,Caspase-3在外阴鳞癌中表达阳性率为53.3%,Ki67的表达与外阴鳞癌的临床病理特征无关(P>0.05),Caspase-3的表达随外阴鳞癌的分化程度降低而降低(P<0.05),Caspase-3蛋白和Ki67蛋白在30例外阴鳞癌中的表达呈负相关(P<0.05)。结论:Caspase-3和Ki67以负反馈的形式参与了外阴癌变的过程,细胞凋亡受阻与细胞增殖异常活跃和外阴癌的发生密切相关。

Expression and correlation of K167 and caspase-3 in vulvar squamous cell carcinoma

Objective:To investigate the expression and clinical significance and correlation of Ki67 and Caspase-3 staining in normal vulvar epithelia and squamous cell carcinomas of the vulvar(VSCC).Methods:The clinical materials of 30 cases of vulvar squamous carcinoma and 10 cases of normal vulvar skin were collected. In our study , immunohistochemistry was employed with S-P method. The immunohistochemistry method employed Mouse anti-human Ki67 monoclonal antibody and rabbit anti-human Caspase-3 polyclonal antibody.Results:The expression of Ki67 in VSCC was 83.3% and the expression of Caspase-3 was 53.3%. The expression of Ki67in VSCC was not related with age ,clinical stage, lymph node metastases or the pathological grade(P>0.05). There was a significant related with the expression of caspase-3 in VSCC and the cell dedifferentiation(P<0.05). The positive expression of Ki67 in VSCC was inversely with caspase-3 (P<0.05).Conclusion:The expression of Ki67 and caspase-3 is inversely correlated in VSCC, the occurrence of VSCC may be related with cell apoptosis inhibition and enhancement of proliferation.
