摘    要:
目的建立HPLC法同时测定外感风痧颗粒(苍耳草、狗仔花、山芝麻等)中原儿茶酸、新绿原酸、原儿茶醛、绿原酸、隐绿原酸、咖啡酸、异绿原酸B、异绿原酸A、异绿原酸C的含有量。方法该药物甲醇提取液的分析采用Hypersil C18色谱柱(250 mm×4.6 mm,5μm);流动相乙腈-0.2%磷酸,梯度洗脱;体积流量1.0 mL/min;柱温25℃;检测波长273、327 nm。对10批样品进行聚类分析、主成分分析。结果 9种成分在各自范围内线性关系良好(r>0.999 0),平均加样回收率95.6%~103.5%,RSD 1.9%~2.8%。10批样品被聚为3类,前2个主成分累积贡献率达91.936%。结论该方法准确、灵敏、可靠,重复性好,可用于外感风痧颗粒的质量控制。

Abstract:
AIM To establish an HPLC method for the simultaneous content determination of protocatechuic acid,neochlorogenic acid,protocatechuic aldehyde,chlorogenic acid,cryptochlorogenic acid,caffeic acid,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C in Waigan Fengsha Granules[Xanthium sibiricum Patrin ex Widder,Vernonia patula(Ait.)Merr.,Helictercs angustifolia L.,etc.].METHODS The analysis of methanol extract of this drug was performed on a 25℃thermostatic Hypersil C18 column(250 mm×4.6 mm,5μm),with the mobile phase comprising of acetonitrile-0.2%phosphoric acid flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelengths were set at 273,327 nm.Cluster analysis and principal component analysis were conducted in ten batches of samples.RESULTS Nine constituents showed good linear relationships within their own ranges(r>0.9990),whose average recoveries were 95.6%-103.5%with the RSDs of 1.9%-2.8%.Ten batches of samples were clustered into three categories,the former two principal components demonstrated the accumulative contribution rate of 91.936%%.CONCLUSION This accurate,sensitive,reliable and reproducible method can be used for the quality control of Waigan Fengsha Granules.
