摘 要: | 目的 评价低温心脏保存液HP100XD对猪离体冠状动脉血管平滑肌功能的保存效果。方法 瑞典家猪10只,全身麻醉及全身肝素化后取出心脏,经冠状动脉开口滴注氧和Krebs溶液去除冠状动脉内血液,于手术显微镜下游离前降支远端血管,并切成1mm长的血管环。随机将血管环分为新鲜对照组、24h保存组和48h保存组,新鲜对照组将血管环直接置入器官浴槽内,24h保存组和48h保存组将血管环置入4℃HP100XD溶液中分别保存24、48h。3组采用高钾Krebs溶液和血栓烷A2相似物(U-46619)检测平滑肌的收缩功能,采用罂粟碱诱导血管环的非内皮依赖性舒张反应评价平滑肌的舒张功能。结果 新鲜对照组由高钾Krebs溶液和U-46619诱导的收缩反应值分别为(22.3±3.3)、(16.4±3.6)mN,24h保存组分别为(21.8±3.4)、(16.3±2.3)mN,48h保存组分别为(22.3±3.3)、(17.3±2.2)mN,3组比较差异均无统计学意义(P〉0.05);3组血管平滑肌的最大舒张百分数均为100%。结论 HP100XD溶液可完整保存离体冠状动脉血管平滑肌功能达48h,是一种较好的心脏保存液。

Abstract: | Objective To evaluate the effect of HP100XD on the hypothermie preservation of the smooth muscle function of isolated porcine coronary artery. Methods Ten Swedish domestic pigs were excised the heart after anaesthesia and systemic heparinization. The blood in coronary artery was removed by dripping oxygenated Krebs solution through the lumen of the vessels. Under opertative microscope, the distal part of anterior descending coronary artery was dissected free from the surrounding tissue, and cut into 1-mm long segments, which were randomly divided into fresh control group, 24-hour preservation group and 48-hour preservation group. The vessel segments were stored in organ bath in fresh control group, and in HP100XD solution at 4 ℃ for 24 and 48 hours in 24- and 48-hour preservation groups. The contraction function of smooth muscle was detected by high potassium Krehs solution and the thromhoxane A2 analogue U-46619, and the relaxation function was detected by endothelium-independent relaxation induced by papaverine immediately. Results The contraction values of smooth muscle induced by high potassium Krebs solution and the thromhoxane A2 analogue U-46619 showed no significant differences among fresh control group ((22.3 ± 3.3), (16.4 ± 3.6) mN), 24-hour preservation group ((21.8± 3.4), (16.3± 2.3) mN) and 48-hour preservation group ((22. 3 ± 3.3), (17.3±2.2) raN) (P〉0.05). The maximal relaxation percentage of smooth muscle was 100% in three groups respectively. Conclusion The smooth muscle of isolated coronary artery could he completely preserved functioning for 48 hours in HP100XD solution, which is a promising solution for preserving heart. |
