青蒿琥酯逆转SHI-1细胞株多药耐药的初步研究

摘    要:目的研究青蒿琥酯逆转人单核细胞白血病SHI-1细胞株多药耐药的效果。方法四甲基偶氮唑盐(MTT)法检测SHI-1细胞株对柔红霉素(DNR)、去甲柔红霉素(ID)、阿糖胞苷(Ara-c)、柔红霉素联合阿糖胞苷(DA)、去甲柔红霉素联合阿糖胞苷(IDA)的敏感性及青蒿琥酯逆转耐药的效果。结果 DA、IDA方案与单用化疗药相比,药物敏感性有提高,且差异具有统计学意义(P〈0.05),非细胞毒性青蒿琥酯联合应用与单用化疗药、DA、IDA方案IC50差异具有显著统计学意义(P〈0.001),随青蒿琥酯浓度的增加,逆转倍数呈上升趋势。结论联合应用非细胞毒剂量青蒿琥酯逆转SHI-1细胞株多药耐药作用显著,明显优于DA、IDA方案。

Preliminary study on reversing of multidrug resistance of SHI-1 cell line by artesunate

Abstract:Aim To investigate the reversal effect of artesunate on drug resistance of SHI-1 cell line. Methods MTT assay was used to detect the sensitivity of SHI-1 cell line to DNR,ID,Ara-C alone and combination of DNR with Ara-C (DA),ID with Ara-C (IDA) and the reversal effect of artesunate on the drugs mentioned above. Results The difference in using DNR,ID,Ara-C alone and combination of DNR with Ara-C(DA),ID with Ara-C(IDA)was significant (P0.05)(There was significantly different between using DNR,ID,Ara-C alone as well as DA,IDA in combination with non-cytotoxicity artesunate at different concentrations (P〈0.001). the reversal impact raised along with the increasing concentration of artesunate. Conclusion The effect of reversal multidrug resistance of non-cytotoxicity artesunate on SHI-1 cell line is significant,and is better than the regimen of DA and IDA.
