健脾止动汤对多发性抽动症模型鼠纹状体多巴胺通路的影响 |

摘 要: | 目的研究健脾止动汤对多发性抽动症(TS)模型鼠纹状体多巴胺通路的影响,为健脾止动汤干预小儿TS提供神经生化学依据。方法90只雄性ICR小鼠随机分成6组,即空白组、模型组、泰必利组、健脾止动汤组、六君子汤组、泻青汤组。除空白组外,其余5组小鼠腹腔注射亚氨基二丙腈建立TS小鼠模型。除空白组和模型组给予生理盐水外,其余4组分别灌胃给予相应药物。4周后采用高效液相色谱电化学法测定TS模型鼠脑纹状体单胺类递质的含量。结果TS模型鼠表现出抽动症特征性的运动行为和刻板行为。小鼠脑纹状体单胺类递质含量,模型组较空白组脑多巴胺(DA)、二羟苯乙酸(DOPAC)、高香草酸(HVA)、5-羟色胺(5-HT)、去甲肾上腺素(NB)含量无明显变化(P〉0.05):泻青汤组较模型组DA、DOPAC含量降低(P〈0.05),泻青汤组较泰必利组脑DA、5-羟吲哚乙酸(5-HIAA)、NE含量降低(P〈0.01或P〈0.05),泻青汤组较六君子汤组脑DA、5-HIAA、NE含量降低(P〈0.01或P〈0.05)。结论健脾止动汤的主要作用组分为泻青汤,其抗抽动作用可能与兴奋脑纹状体多巴胺间接通路、降低DA含量有关。

Abstract: | Objective To study the effects of Jianpizhidong decoction on dopamine pathway in TS model mice striatum, and provide neural biochemical proof for Jianpizhidong decoction in treating children Tourette syndrome (TS). Methods 90 male ICR mice were randomly divided into 6 groups, namely blank control group, model group, Tiapride group, Jianpizhidong decoction group, Liujunzi group, Xieqing group. Except blank control group, other 5 groups mice were made to TS models by intraperitoneal injection of 3, 3'-Iminodipropionitrile (IDPN). With the exception of blank and model groups, other 4 groups were given corresponding drugs by intragastric administration respectively. 4 weeks later, the levels of monoarnine neurotransmitter in brain striatum of TS model mice were detected by high performance liquid chromatography and electrochemical detection. Results TS model mice appeared characteristic locomotion behavior and stereotypy behavior. The levels of monoamine neurotransmitters with the exception of 5-HIAA had no difference in striatum of mice between TS model group and blank control group (P〉0.05). Compared with model group, levels of DA and DOPAC in striatum reduced in Xieqing group (P〈0.05). Compared with Tiapride group, levels of DA, 5-HIAA, NE in striatum reduced in Xieqing group (P〈0.01, P 〈0.01, P 〈0.05). Compared with Liujunzi group, levels of DA, 5-HIAA, NE in striatum reduced in xieqing group (P 〈0.01, P 〈0.01, P 〈0.05). Conclusions The mainly component of Jianpizhidong decoction is Xieqing decoction, the function of improving symptoms of TS maybe relate to activate indirect dopamine pathway and decrease levels of DA in TS model mice striatum. |
