大鼠慢性溃疡性结肠炎模型建立方法探讨

摘    要:目的:探讨TNBS/乙醇法诱导大鼠溃疡性结肠炎模型溃疡、炎症持续时间及如何建立持续而稳定的大鼠慢性溃疡性结肠炎模型。方法:SD大鼠70只,随机分成空白组(10只)、模型A组和模型B组(各30只)。除空白组外各模型组采用TNBS/乙醇法诱导大鼠溃疡性结肠炎模型,模型A组大鼠给予一次性灌肠造模;模型B组每隔7d重复造模,持续至第8周。各模型组大鼠分别于造模第2周、5周、8周(空白组大鼠于造模第8周)随机抽取8只大鼠处死,进行结肠病变肉眼评分及结肠组织病理学评分。结果:与空白组比较,模型A组大鼠的体重、结肠病变肉眼评分及病理学评分在第2周有显著性差异(P〈0.01),在第5周、第8周无差异(P〉0.05);模型B组大鼠的体重、结肠病变肉眼评分及病理学评分在第2周、第5周、第8周均有差异或显著性差异(P〈0.05或P〈0.01)。结论:采用TNBS/乙醇法每周灌肠造模可成功构建持续而稳定的大鼠慢性溃疡性结肠炎模型。

Abstract:Objective : To discuss the ulceration and inflammation duration of a rat model of 2,4,6- trinitrobenzenesulfonie acid ( TNBS )/ethanol-induced ulcerative colitis and how to build sustainable and stable rat model of chronic ulcerative colitis. Method : Seventy SD rats were divided randomly into 3 groups : normal group, model groups A and B.Model groups A and B were used TNBS/ethanol to induce rat model of ulcerative colitis.Rats of model group A were given enema at a time while model group B were given enema every other 7 days to repeat molding, continued to eighth weeks.8 rats of model groups A and B were killed in second week, fifth week and eighth week after model making, then colonic lesions gross score and colon tissue pathology score were evaluated. Results: Compared with normal group, the weight, colonic lesions gross score and colon tissue pathology score of model group A were significantly different in second week (P〉0.01) and had no significant difference in fifth week and eighth week (P〉0.05)after model making. Model group B were significantly difference in second week, fifth week and eighth week ( P〈0.05 or P〈0.01 ) after model making. Conclusion : Using TNBS/ethanol method every week can successfully create sustainable and stable rat model of chronic ulcerative colitis.
