LLN对大鼠急性心肌损伤保护作用研究

摘    要:目的:研究LLN对大鼠急性心肌损伤的保护作用及其部分机制。方法:用异丙肾上腺素造成大鼠心肌损伤模型。SD大鼠随机分成6组:正常组、ISO模型组(30mg/kg)、LLN大剂量组(400mg/kg)、LLN中剂量组(200mg/kg)、LLN小剂量组(100mg/kg),丹参组(400mg/kg),镜下观察药物对急性心肌损伤大鼠心肌组织病理形态学的影响;免疫组化检测对大鼠心肌细胞bcl2、bax、caspase3蛋白表达的影响。结果:ISO组心肌坏死明显,bax、caspase3蛋白表达增加;LLN组和丹参组心肌坏死减轻,bax、caspase3表达下降,bcl2/bax升高。结论:LLN对异丙肾上腺素造成的大鼠急性心肌损伤有保护作用,其机理与升高bcl2/bax比值,降低caspase3的表达和抑制凋亡有关。

Abstract:Objecive: To study the protection effect and possible mechanisms of LLN in acute myocardial injury induced by isoprenaline (ISO) in rats. Methods: Acute myocardial injury was induced by hypodermic injecting ISO 30mg/kg. SD rats were divided into six groups at random.They were control group,the ISO group,the LLN group(400mg/kg), the LLN group(200mg/kg), the LLN group(100mg/kg)and the Danshen group (400mg/kg);The effect of LLN in myocardium was observed by pathomorphism. The express of bcl-2,bax,caspase-3 was studied by ABC immunohistochemistry. Results: In the ISO group,the myocardial necrosis were more serious.Moreover the express of bax,caspase-3 increased.In the LLN group and the Danshen group,the myocardial necrosis were relieved. the The express of bax,caspase-3 decreased,bcl-2/bax increased. Conclusion: The LLN can alleviate rat myocardium from injury by increasing bcl-2/bax,decreasing the expression of caspase-3 and reducing myocardial apoptosis.
