摘    要:目的:探讨PI3K/AKT和MAPK/ERK1/2信号通路对人胰腺癌PANC-1血管内皮生长因子(VEGF)表达的影响。方法:抑制剂LY294002和PD98095分别处理人胰腺癌PANC-1细胞,Western blotting检测细胞VEGF蛋白表达。结果:分析表明LY294002和PD98095处理后的PANC-1细胞的磷酸化AKT、磷酸化ERK1/2及VEGF蛋白表达水平均明显低于对照组VEGF(P<0.05)。结论:抑制PI3K/AKT和MAPK/ERK1/2细胞信号通路能够下调VEGF蛋白表达,进而抑制肿瘤新生血管的生成及浸润转移能力。

Abstract:Objective:To investigate effects of PI3K/AKT and MAPK/ERK1/2 signaling pathways on human pancreatic cancer PANC-1 cells and expression of vascular endothelial growth factor(VEGF).Methods:PANC-1 cells,Western blotting testing cells and protein expression of VEGF were exposed to inhibitor LY294002 and PD98059 respectively.Results:The analysis shows that the level of phosphorylation of AKT and ERK in PANC-1 cells and the protein expression of VEGF disposed to inhibitor LY294002 and PD98059 is significantly lower than the control group(P<0.05).Conclusion:Inhibiting the PI3K/AKT and MAPK/ERK1/2 signaling pathways can regulate the protein expression of VEGF.Furthermore,it inhibits the production,invasion and metastasis ability of tumor neovascularization.
