摘    要:目的 建立测定人血浆中地西泮和咪达唑仑质量浓度的高效液相色谱法.方法 色谱柱采用Waters C18柱(250 mm x4.6mm,5μm),流动相为乙腈-水(55∶45),柱温30℃,流速1.0 mL/min,检测波长220 nnl.结果 地西泮和咪迭唑仑的最低检测质量浓度为5 ng/mL,进样质量浓度的线性范围为O.05~10.0μg/mL(r>0.990 0);咪达唑仑和地西泮日内精密度的RSD分别为3.09%~5.33%和1.56%~4.25%,日间精密度的JRSD分别为4.72%~6.52%和2.59%~6.22%,方法回收率分别为94.02%~97.19%和96.07%~105.64%.结论 所用方法简便、快速、准确,适用于地西泮和咪达唑仑体内血药浓度和生物等效性的测定.

Abstract:Objective To determine diazepam and midazolam in human plasma by HPLC.Methods The HPLC separation was carried out on a Waters C18 column(250 mm×4.6 mm,5 μm),using a UV detector and the wavelength was 220 nm,and mobile phase was acetonitrile-water(55∶45).Results For diazepam and midazolam,the limits of detection were 5 ng/mL.The concentration of midazolam and diazepam showed a linear plot in the range of 0.05-10.0 μg/mL(r 0.990 0).Within-day and between-day precisions expressed as the relative standard deviation(RSD) for the method were 3.09%-5.33%,1.56%-4.25%,4.72%-6.52%,and 2.59%-6.22%,respectively.The extraction recoveries were 94.02%-97.19%and 96.07%-105.64%,respectively.Conclusion The method is proved to be accurate,sensitive and reliable.It is suitable for the simultaneous determination in human plasma of diazepam and midazolam.
