MRL/lpr小鼠中CD4+CD25+调节性T细胞含量、增殖能力及抑制能力的研究

摘    要:目的探讨MRL/lpr小鼠中CD4+CD25+调节性T细胞(Treg细胞)的含量、增殖能力及其对效应性T淋巴细胞(Teff细胞)的抑制能力及意义。方法流式细胞检测法检测8只MRL/lpr小鼠及8只BALB/c小鼠脾脏中Treg细胞的水平;磁珠分选法分选出脾脏中的Treg细胞与Teff细胞,离体培养2周后,分别测量Treg细胞数量,并比较差异;用Treg细胞与Teff细胞组成共培养体系,5 d后检测Teff细胞的水平,计算Treg细胞对Teff细胞的抑制率并比较差异。结果 MRL/lpr小鼠脾脏中Treg细胞的总含量与BALB/c小鼠无统计学差异(4.88±0.55)%vs(5.03±0.34)%,P>0.05];离体培养2周后MRL/lpr小鼠的Treg细胞数量显著低于BALB/c小鼠(6.61±0.92)×105vs(13.25±1.91)×105,P<0.05];共培养5 d后MRL/lpr小鼠Treg细胞对Teff细胞的抑制率显著低于BALB/c小鼠Treg细胞(P<0.05)。结论 MRL/lpr小鼠体内Treg细胞增殖能力、抑制能力的下降可能导致体内免疫平衡的破坏、自身抗原-抗体反应,参与MRL...
