卡介苗激活系统血液快速固有免疫反应的体外仿真实验研究

摘    要:目的对卡介苗激活正常人与肺癌患者系统血液快速固有免疫反应水平进行比较研究。方法将卡介苗(BCG 1mg·mL^-1)0.2mL(实验组)或生理盐水0.2mL(对照组)加入枸橼酸抗凝全血细胞悬液0.2mL和新鲜血浆0.3mL中,37℃温育1h.用流式细胞仪测定各试管反应液中红细胞CD59和淋巴细胞CD25平均荧光强度。结果正常人实验组红细胞CD59分子表达水平(32.60±3.15)高于正常人对照组红细胞CD59分子表达水平(29.96±5.40),高于肺癌实验组红细胞CD59分子表达水平(28.69±2.88)和肺癌对照组红细胞CD59分子表达水平(28.36±4.69),P〈0.05;正常人实验组淋巴细胞CD25分子表达水平(5.99±1.78)高于正常人对照组淋巴细胞CD25分子表达水平(5.43±1.21)。高于肺癌实验组淋巴细胞CD25分子表达水平(5.28±1.78)和肺癌对照组淋巴细胞CD25分子表达水平(4.60±1.38),P〈0.05。结论卡介苗能激活系统血液快速固有免疫反应,促进红细胞CD59分子和淋巴细胞CD25分子表达,但肺癌患者卡介苗促进红细胞CD59分子表达的激活能力明显低于正常人中对红细胞CD59分子的表达量的激活能力。

Abstract:Objective To compare the activation level of Bacille Calmette Guerin to systemic hematogenic quick innate immunoreaction between normal people and patients with lung cancer. Methods Bacille Calmette Guerion(BCG 1 mg· mL^-1 ) 0.2 mL (an experimental group) or NS (a control group) 0.2 mL was added to anticoagulant suspension whole blood cells 0.3 mL, and then incubated for 1 h at 37℃. The average fluorescence intensity for CD59 on red blood cells and CD25 on lymphocytes was determined by flow cytometer. Results The level of CD59 on red blood cells in the experimental group of normal people (32.60 ± 3. 15) was significantly higher than that in the contrastive group of normal people (29.96±5.40), that of CD59 on red blood cells in the experimental group of lung cancer (28.69±2.88), and that on red blood cells in contrastive group of lung cancer (28.36±4.69) (P〈0. 05). The level of CD25 on lymphocytes in the experimental group of normal people (5.99±1.78) was significantly higher than that of CD25 on lymphocytes in the experimental group of lung cancer (5.43± 1.21), that of CI)25 on lymphocytes in the contrastive group of normal people (5.28±1.78), and that of CD25 on lymphocyte in the contrast ire group of lung cancer (4.60 ±1.38) (P〈0.05). Conclusion Bacille Calmette (;uerin can activate systemic hematogenic quick innate immunoreaction to enhance the expression of CD59 on red blood cells and CD25 on lymphocytcs. But the activation level of Bacille Calmette Guerin to enhance the expression of CD59 on red blood cells in lung cancer significantly is lower than that of CD59 on red blood cells in normal people.
