外源性可溶性促红细胞生成素受体对正常大鼠视网膜神经元存活的影响

摘    要:
目的观察玻璃体腔注射外源性可溶性促红细胞生成素受体(EPOsR)对视网膜神经元存活的影响。方法30只SD鼠随机分为正常对照组、PBS组、EPOsR 2ng组、EPOsR 20ng组、EPOsR 200ng组,每组6只(12只眼)。后4组玻璃体腔内分别注射5μL PBS,2、20、200ng EPOsR。注射前记录闪光视网膜电图(FERG),注射后3d再次进行FERG检查及视网膜神经元TUNEL凋亡原位检测,注射后7d进行光学显微镜和透射电镜检查。结果注药前后各组a波、b波的潜伏期及振幅差异无统计学意义(P〉0.05),震荡电位(OPs)的P4潜伏期和各子波总振幅差异亦无统计学意义(P〉0.05);全层视网膜神经元TUNEL凋亡原位检测未见凋亡细胞;光学显微镜和透射电镜检查均未见明显的水肿、空泡状变性等组织病理学改变。结论玻璃体腔注射200ng以下剂量的外源性EPOsR不影响正常状态下视网膜神经元的存活。

Abstract:
Objective Previous studies have shown that erythropoietin soluble receptor (EPOsR) can exacerbate retinal ischemic damage,but it remains unclear whether EPOsR can affect the survival of normal retinal neurons. The aim of the present study was to investigate the effect of EPOsR on the survival of normal retinal neurons. Methods Thirty albino Sprague-Dawley (SD) rats were evenly randomized into normal control group,PBS group and EPOsR 2 ng,20 ng,200 ng groups and 6 rats (12 eyes) for each. PBS or EPOsR 2,20,200 ng was injected into the vitreous of rabbits. Flash electroretinogram (FERG) was recorded before intravitreal injection and repeated injection at 3 days. At the same time-point,3 rats of each group were sacrificed for apoptosis assay. Seven days after intravitreal injection,another 3 rats were sacrificed for histopathological examination. One eyeball of each rat was for light microscopy and the another for transmission electromicroscopy. Results Compared with preinjection,the latencies and amplitudes of FERG a- and b-wave in postinjection were not changed significantly (P>0.05). There was no significant difference in OPs latency and total amplitude between preinjection and postinjection (P>0.05). No apoptotic cells were presented within retinas in any group after injection,and no obvious edema or vacuolar degeneration was observed in retinas in any group under light microscope and transmission electromicroscope. Conclusion Intravitreal administration of EPOsR at dose of 200 ng does not affect the survival of normal retinal neurons.
