Effect of antimicrobial peptide LL-37 on proliferation,invasion and apoptosis of SCC-090 cells in oral squamous cell carcinoma

Objective To study the effect of antimicrobial peptide LL-37 on the proliferation,invasion and apoptosis of SCC-090 cells in oral squamous cell carcinoma.Methods Oral squamous cell carcinoma SCC-090 cells were cultured in vitro,the blank control group(antibacterial peptide LL-370μg/mL),and different concentrations of antibacterial peptide LL-37(25,50,100,150,200μg/mL)were setted,after culturing the cells for 24,48,and 72 hours,the proliferation of the cells was tested by the cell proliferation-toxicity method,the effect of LL-37 on OSCC cell invasion and migration was detected by transwell assay,and apoptosis was determined by Annexin V/PI double staining.The effect of LL-37 on the secretion of tumor necrosis factorα(TNF-α),interleukin-6(IL-6),and interleukin-8(IL-8)content was tested by enzyme-linked immunosorbent assay(ELISA)kit.Results Compared with the control group,the proliferation of SCC-090 cells was inhibited by adding different concentrations of antibacterial peptide LL-37(25,50,100,150,200μg/mL),and the degree of inhibition increased with the increase of LL-37 concentration and time,the difference was statistically significant(P<0.05).With the increase of LL-37 concentration,the cell apoptosis rate gradually increased,and the cell invasion ability weakened,the difference was statistically significant(P<0.05).With the increase of the concentration of LL-37,the contents of TNF-α,IL-6,and IL-8 in the cell supernatant decreased,and the difference was statistically significant(P<0.05).Conclusion Antimicrobial peptide LL-37 can inhibit the proliferation and invasion of oral squamous cell carcinoma cells in a dose-dependent manner,promote the apoptosis of cancer cells,and inhibit the secretion of TNF-α,IL-6,and IL-8.
