单纯疱疹病毒Ⅱ型在Vero细胞中培养与增殖特性的研究

目的研究单纯疱疹病毒Ⅱ型（HSV-2）在非洲绿猴肾细胞（Vero细胞）中培养及增殖的合适条件。方法把HSV-2接种于Vero细胞中培养和传代,于不同时间收获病毒液并测毒力,进行不同培养条件的摸索。结果pH值、残留牛血清是HSV-2在Vero细胞培养的影响因素,最佳收毒时间为36-72h。在Vero细胞上培养病毒3-5代,病毒毒力较高。结论建立了单纯疱疹病毒Ⅱ型在Vero细胞上培养增殖的初步方法,为下一步建立HSV-2的潜伏与激发细胞模型打下基础。

Incubation of Herpes Simplex virus type 2 in Vero cells and its proliferating features

Objective To study the appropriate incubation and proliferation conditions of Herpes Simplex virus type 2 （HSV-2） in Veto cells from African green monkeys. Methods HSV-2 was cultured and sub-cultured after inoculated into Vero cells. Virus suspension was harvested at different time points and titration of virus was tested. The proliferation condition of HSV-2 was investigated. Results The pH value and residual calf serum were the influencing factor for the proliferation of HSV-2 in Vero cells. The optimal time to harvest the virus was 36-72 hours after inoculation. The toxicity of HSV-2 was increased after 3-5 passages of HSV-2 in Vero cells and reached its peak after 10 passages. Conclusion The incubation and proliferation procedure for HSV-2 has been established, which lays a foundation for the establishment of HSV-2 latency and reactivation in vitro.
