摘    要:目的探讨适宜的玻璃酸钠中蛋白质含量测定方法。方法用Lowry改良的酚试剂法和Bradford染料结合法分别测定玻璃酸钠中和酶解前后牛血清白蛋白溶液中蛋白质的含量。结果玻璃酸钠的蛋白质含量用Lowry法测得结果偏高 ;白蛋白溶液酶解前两种方法测得结果相当 ,酶解后Lowry法所测值略有上升 ,而Bradford法测定值却大幅下降。结论Lowry法测定蛋白质含量 ,小分子多肽的干扰较大 ,使得结果偏高 ;Bradford法更适合于测定玻璃酸钠中微量蛋白质的含量

Abstract:PurposeTo select the suitable method for the protein determination in sodium hyaluronate.MethodsThe differences between Folin phenol method modified by Lowry and Bradford′s dye binding method for determination of protein in sodium hyaluronate, and in bovine serum albumin (BSA) solution before and after enzymolysis were compared.ResultsValues of protein content in sodium hyaluronate determined by Lowry′s methods are much higher than Bradford′s method. Before hydrolysis, the values of protein content in BSA solution determined by the two methods are similar. But after hydolysis, the values determined by Lowry′s method become slight higher, on the contrary, the values determined by Bradford′s method become much lower.ConclusionLowry′s method is interfered by peptide, so the values are higher. Bradford′s method is more suitable for determination of protein in sodium hyaluronate.
