硫磺熏蒸前后金银花的HPLC指纹图谱比较和聚类分析

摘    要:目的:建立硫磺熏蒸前后金银花HPLC指纹图谱,为评价硫磺熏蒸前后金银花样品的质量优劣提供依据。方法:采用RP-HPLC方法,色谱柱:Pursuit XRs C18(250 mm×4.6 mm,5μm);流动相:乙腈-0.1%磷酸水溶液,梯度洗脱;柱温30℃;检测波长238 nm。结果:建立了硫磺熏蒸前后金银花HPLC指纹图谱共有模式,并指认了其中4个色谱峰。通过与对照品的保留时间比较,4,5,13,14号峰的归属分别为绿原酸、咖啡酸、芦丁、木犀草苷。通过采用相似度计算法结合聚类分析对19批金银花进行比较分析,从而对其质量进行评价。结论:金银花在硫磺熏蒸前后,某些共有成分的含量会有不同程度的改变,硫磺熏蒸对金银花的质量影响较大。

Abstract:Objective: To establish the chromatographic fingerprint of lonicerae japonicae flos before and after sulfur-fumigated process,and to provide evidence for evaluating the qualities of lonicerae japonicae flos samples.Methods: RP-HPLC method was applied to establish the chromatographic fingerprint.The separation was performed on a Pursuit XRs C18 column with gradient elution of mobile phase composed of acetonitrile-aqueous solution containing 0.1% phosphoric acid.The column temperature was 30 ℃ and the detective wavelength was set at 238 nm.Results: The common pattern of chromatographic fingerprint of lonicerae japonicae flos before and after sulfur-fumigated process was established,and 4 peaks of them were identified.The 4th,5th,13th and 14th peaks were assigned to chlorogenic acid,caffeic acid,rutin and galuteolin respectively by comparison with the retention times and UV spectra of reference substances.The qualities of 19 batches of lonicerae japonicae flos samples were compared and evaluated by using similarity algorithm combined with clustering analysis.Conclusion: The contents of many common peaks of lonicerae japonicae flos were changed at different degrees before and after sulfur-fumigated process.Sulfur-fumigated process may have great influence on the quality of lonicerae japonicae flos.
