Abstract:Objective It is to study the effects of electroacupuncture at different frequencies on neuroglobin(Ngb)and NLRP3 signaling pathway related indicators in hematoma brain tissue of rats with intracerebral hemorrhage.Methods Ninety rats were randomly divided into six groups:normal group,sham operation group,model group,electroacupuncture group I,electroacupuncture group II and electroacupuncture group III,each group had 15 rats.The cerebral hemorrhage models were replicated by autologous arterial blood infusion into the caudate nucleus of rats in the model group and the electroacupuncture group.After the rats were awakened from the modeling,the ones in each electroacupuncture group were treated with selected electroacupuncture stimulation at Baihui,Shuigou,and bilateral Neiguan.The electroacupuncture frequency was respectively set for 2 Hz,15 Hz,and 50 Hz in group I,group II and group III,once a day,totally continuously treated for 10 days;the rats in the sham operation group were treated with telescopic relief needle,but the skin was not penetrated.The scores of the neurobehavior of rats in each group were evaluated by Bederson scoring standard,the brain tissue morphology was observed by HE staining,the expression of Ngb in hematoma brain tissue was detected by Western blot,and the contents of NLRP3,Caspase-1 and interleukin-1β(IL-1β),interleukin-18(IL-18)in hematoma brain tissue were detected by ELISA method.Results At 24h,48h,5d and 10d in the study,the neurobehavioral scores of each electroacupuncture group were significantly lower than those of the model group(all P<0.05),and the scores showed a significant downward trend as time passed and the frequency of electroacupuncture increased.The neurobehavioral scores of the electroacupuncture group III at 5 d and 10 d were significantly lower than those of the electroacupuncture group I(all P<0.05).HE staining showed that the cells in the cerebral hemorrhage area of the model group were irregularly arranged,a large number of degenerated and necrotic nerve cells were found,the intercellular space became larger,the cytoplasm was reduced,and most of the nucleus pyknosis was found.The cell morphology of the cerebral hemorrhage area of rats in each group of electroacupuncture was improved compared with the model group,and the improvement of electroacupuncture group III was the most obvious.The expression of Ngb in hematoma brain tissues in each group of electroacupuncture was significantly higher than that of the model group(all P<0.05),and both electroacupuncture group II and electroacupuncture group III were significantly higher than electroacupuncture group I(all P<0.05).The contents of NLRP3,Caspase-1,IL-1β,and IL-18 in the hematoma brain tissue of each group of acupuncture were significantly lower than those of the model group(all P<0.05),and they showed a gradual decline with the increase of the frequency of electroacupuncture.Conclusion Electroacupuncture of different frequencies can affect the therapeutic effect of cerebral hemorrhage rats.Among them,50 Hz high-frequency electroacupuncture has a better effect than 2 Hz and 15 Hz low-frequency electroacupuncture.The therapeutic effect may be achieved by regulating the expression of Ngb in hematoma brain tissue and the NLRP3 signaling pathway.
