脑血疏口服液对脑出血大鼠神经功能及CD36表达的影响

摘    要:目的观察脑血疏口服液对脑出血大鼠神经功能及脑组织CD36表达的影响。方法采用Ⅶ型胶原酶以大鼠尾状核为目的注射点制备大鼠脑出血模型。采用随机数字表法将无特定病原体动物(SPF)大鼠分为假手术组、模型组、脑血疏组,每组6只。脑血疏组按照3.6 mL/kg灌胃给药,假手术组及模型组分别给予等量生理盐水。造模后连续3 d模型组和脑血疏组采用改良的神经功能缺损评分标准(mNSS)进行神经功能缺损评分,苏木精-伊红染色法(HE)观察大鼠血肿周围组织病理形态学改变,免疫组化观察CD36阳性细胞数。蛋白质印迹法(Westem Blot)检测各组血肿周围组织CD36的表达变化。结果模型组与脑血疏组大鼠均出现不同程度的神经功能缺失症状,脑血疏组神经功能恢复较模型组快;与模型组比较,脑血疏组HE染色坏死面积减少,周围组织疏松减轻,神经细胞核结构较清晰。Westem Blot检测脑血疏组CD36表达量高于模型,免疫组化检测脑血疏组血肿周围CD36阳性细胞数高于模型组。结论脑血疏口服液可以通过增加大鼠脑出血后CD36表达,改善其神经功能缺失及促进血肿吸收,减轻脑出血后继发脑损害。

Abstract:Objective To observe the effect of Naoxueshu oral liquid on neurological function and CD36 in rats with intracerebral hemorrhage.Methods The model of intracerebral hemorrhage in rats was established by injection point of typeⅦcollagenase in the caudate nucleus of rats.Specific pathogen free(SPF)rats were randomly divided into sham operation group,model group and cerebral hemopolyxia group,with 6 rats in each group.The cerebral hemopolyxia group was gavaged 3.6 mL/kg,and the sham operation group and the model group were gavaged the same normal saline.After 3 days of modeling,Modified Neurological Severity Score(mNSS)was used to evaluate the neurological deficit in the model group and the cerebral hemopolyxia group.At the fourth day of modeling,the pathological morphological changes of the tissues around the hematoma were observed by HE staining,the number of CD36 positive cells was observed by immunohistochemistry,and the expression of CD36 in the brain tissues around the hematoma in each group was detected by Western Blotting.Results Different symptoms of neurological function loss appeared in model group and cerebral hemopolyxia group,and neurological function recovery in cerebral hemopolyxia group was faster than that in model group.Compared with model group,hematoxylin-eosin(HE)staining showed that necrotic area decreased and peripheral tissue loosened,and the structure of neuronal nucleus was clearer in cerebral hemopolyxia group.The expression of CD36 in cerebral hemopolyxia group was higher than that in model group by Western blotting.The number of CD36 positive cells around hematoma in cerebral hemopolyxia group was more than that in the model group.Conclusion Naoxueshu oral liquid could improve the expression of CD36 in rats after intracerebral hemorrhage,and the neurological function,promote the absorptionof hematoma,and reduce the secondary brain damage after intracerebral hemorrhage.
