Abstract:Objective:To investigate the effects of hsa_circ_0003188 on osteogenic differentiation of human bone marrow mesenchymal stem cells(hBMSCs).Methods:Human Circular RNA Microarray Version 2.0 microarray was used to detect the differentially expressed circRNAs in hBMSCs cultured in normal and inflammatory environment(10 ng/mL TNF-α)respectively and to screen out the target circRNA-hsa_circ_0003188.After Lent-hsa_circ_0003188 transfection into hBMSCs and osteogenic induction,the effect of high expression of hsa_circ_0003188 on osteogenic differentiation of hBMSCs was detected by alkaline phosphatase staining.TargetScan and other biological softwares were used to predict the biological function of hsa_circ_0003188.Results:hsa_circ_0003188 was highly expressed in hBMSCs in inflammatory environment and Lent-hsa_circ_0003188 inhibited the osteogenic differentiation of hBMSCs.Biology softwares predicted the ceRNA mechanism of hsa_circ_0003188.Conclusion:hsa_circ_0003188 can inhibit the osteogenic differentiation of hBMSCs,which may become a new regulatory target for hBMSCs during bone regeneration.
