MMP-2和PTEN蛋白在肝癌组织中的表达及临床意义

目的 探讨肝癌组织中MMP-2和PTEN的表达及两者与肝癌侵袭性的关系.方法 用免疫组化SP法检测61例肝癌组织和17例正常肝组织中MMP-2和PTEN蛋白质的表达,并分析两者与肝癌临床病理特征,侵袭性的关系.结果 HCC中PTEN阳性表达率明显弱于正常肝组织,而MMP-2阳性表达率明显高于正常肝组织,且两者呈负相关;PTEN蛋白阳性表达率与HCC组织分化程度、侵袭性有关(P＜0.05),而与性别、术前AFP水平无关(P＞0.05);MMP-2阳性表达率与HCC组织侵袭性有关(P＞0.05),而与HCC组织分化程度、性别、术前AFP水平无关(P＜0.05).结论 PTEN、MMP-2的表达一定程度上反映HCC侵袭性强弱;PTEN缺失可引起MMP-2表达增加,在HCC侵袭、转移中发挥作用.

Clinical Study of Relationship between Expression of MMP-2 and PTEN in Hepatocellular Carcinoma

Objective To explore the relationship of the expressions of MMP-2 and PTEN of its malignant behaviors in hepatocellular carcinoma(HCC).Methods By using S-P method,the expression of PTEN and MMP-2 in 61 cases of HCC and 17 cases of normal liver tissues was detected,and their relationship with clinicopathological characteristics and invasion of HCC was analyzed.Results The positive expression intensity of PTEN in HCC was remarkably weaker than that in normal liver tissues,while the expression intensity of MMP-2 was stronger in HCC than that in normal liver tissues.Moreover,there was a negative relationship between the expression of PTEN and that of MMP-2.The positive expression rate of PTEN in HCC was related to the progress of the tumor pathological differentiation and invasion(P<0.05),whereas that wasn't related to the sex and AFP levels(P>0.05).The positive expression rate of MMP-2 in HCC was related to the invasion(P<0.05),whereas that didn't show correlation to the tumor pathological differentiation,sex and AFP levels.(P>0.05).Conclusion The expression of PTEN and MMP-2 may reflect the differentiation and invasive potential of HCC tissues to varying degrees.The deletion of PTEN can result in the over expression of MMP-2,which may play an important role in the invasion and metastasis of HCC.
