参桃软肝丸含药血清对HepG-2细胞凋亡及细胞周期影响的实验研究

目的：观察参桃软肝丸含药血清对人肝癌细胞HepG-2细胞凋亡及细胞周期的影响，验证参桃软肝丸可能的作用机理。方法：将9只雄性新西兰家兔分为3组，每组3只，实验分空白对照组、实验组和阳性药物对照组，人肝癌细胞株HepG-2细胞接种于RPMI 1640培养液中，细胞培养为3～5d。MTT法观察细胞抑瘤率，流式细胞仪检测细胞凋亡率和细胞周期。结果：参桃软肝丸对人肝癌细胞株HepG2的生长有抑制作用，且随剂量的增加而增强。其中，高浓度组抑瘤率最高可达43.2%。有较好的诱导凋亡作用，凋亡率可达15.6%，且阻滞细胞增殖于G0/G1期。结论：参桃软肝丸对人肝癌细胞HepG-2有较好的抑瘤和诱导细胞凋亡作用，诱导肿瘤细胞凋亡可能是其抗肿瘤作用机制之一。

Experimental study of the pharmaceutical serum of Shentao Ruangan pills containing serum on the effect of the HepG-2 cells apoptosis and cycle

Objective： To investigate the Shentao Ruangan pills containing serum on the effect of the HepG-2 cells apoptosis and cycle, and to verify the possible action mechanism of the Shentao Ruangan pill. Methods： 9 New-Zealand male rabbit were randomly divided into three groups, treated group, blank serum group and positive group, 3 for each. The human hepatoma cell line HepG-2 cells were seeded in RPMI 1640 culture medium, for 3-5d. Observing the cell inhibition rate in MTT, and detecting apoptosis rate and cell cycle in flow cytometry. Results： Shentao Ruangan pills have inhibitory effect on the growing of the human hepatoma cell line HepG2, and increased with the dose increasing. Among them, the inhibition rate in the high concentration group was up to 43.2%, had a better induction of apoptosis; apoptosis rate reached 15.6%, and block cell proliferation at G0/G1 phase. Conclusion： Shentao Ruangan pills were effective in inhibition and induce apoptosis of the human hepatoma cell line HepG-2, induce tumor cell apoptosis may be one of the mechanisms of its anti-tumor effect.
