摘 要: | 目的采用盲肠结扎穿孔脓毒症模型,研究乌司他丁对脓毒症大鼠血清E-选择素水平和循环内皮细胞的影响。方法SD大鼠72只随机分为3组:A组(假手术组,n=8),B组(脓毒症组,n=32),C组(乌司他丁组,n=32),A组和B组于术后立即皮下注射生理盐水5 mL,C组皮下注射乌司他丁(100 000 U.kg-1,溶于5 mL生理盐水中)。A组术后和B组、C组术后3,6,12和24 h四时相点采血检测血清E-选择素,同时行循环内皮细胞检测。结果 B组和C组各时相点血清E选择素水平及循环内皮细胞计数显著高于A组(P0.01),而各时相点C组血清E选择素水平及循环内皮细胞计数又显著低于B组(P0.05)。结论脓毒症大鼠存在血管内皮细胞的活化及损伤;在脓毒症早期乌司他丁干预可能对内皮细胞有一定保护作用。

Abstract: | Objective: To study the effects of ulinastatin to E-selectin and the number of circulating endothelial cells(CEC) in septic rats dynamicly.Sepsis model induced by cecal ligation and puncture is used.Methods 72 SD rats were randomly assigned into three groups as follows: A(sham group,n = 8);B(sepsis group,n = 32);C(ulinastatin group,n = 32).Group B and group C were further divided respectively into 3,6,12 and 24 h subgroups after operation and each subgroup had 8 rats.In group A B,the animals were subcutaneously injected saline of 5 mL respectively,group C was injected ulinastatin(100 000 U.kg-1,dissolved in normal saline of 5 mL) at the time of operation.The changes of E-selectin,the number of circulating endothelial cells in all rats were measured(rats in sham group were inspected immediately after the operation).Results Compared with group A,the levels of E-selectin and the number of CEC were increased significantly in group B and C(P 0.01).The levels of E-selectin and the number of CEC in group C were significantly lower than those in group B(P 0.05).Conclusions The damage of endotheliocyte was gradually aggravated with the development of sepsis.Ulinastatin used at the early stage may have a certain protective effect on endotheliocyte after sepsis. |
