银杏叶提取物对大鼠局灶性脑缺血再灌注损伤的保护作用

摘    要:目的:研究银杏叶提取物(EGB)对大鼠局灶性脑缺血再灌注损伤的保护作用。方法:采用线栓法制备大鼠大脑中动脉缺血再灌注模型,随机分成假手术、模型、丹参和EGB组,分别在脑缺血再灌注后24h处死大鼠,观察其神经功能缺失评分,应用TTC染色观察梗死体积,尼氏染色观察神经元形态结构特征并计数健存神经元数量,检测血清氧化物歧化酶(SOD)和丙二醛(MDA)变化,Western-blot检测脑组织诱导型一氧化氮合酶(iNOS)表达。结果:经过EGB治疗后,EGB组行为学评分和脑组织梗死体积测量均低于模型组(P<0.05)。EGB组的坏死及凋亡细胞大大减少,健存神经元数目((66.91±7.53)%)较模型组((43.51±4.77)%)显著增多(P<0.05)。EGB组血清SOD高于模型组,MDA低于模型组(P<0.05)。24h后模型组iNOS表达较假手术组和EGB组明显增高(P<0.05)。结论:EGB能显著减小梗死范围,减少神经细胞凋亡,提高SOD含量,降低MDA含量和抑制iNOS表达,对大鼠局灶性脑缺血再灌注损伤具有良好的神经保护作用。

Abstract:OBJECTIVE:To study the protective effect of Ginkgo biloba extracts on focal cerebral ischemia/reperfusion injury in rats.METHODS:The models of ischemia-reperfusion were established by focal middle cerebral artery occlusion(MCAO).The rats were randomly divided into sham operation group,ischemia-reperfusion group,Salvia miltiorrhize.and G.biloba extracts group.Model rats were sacrificed 24 h after ischemia/reperfusion.The neurological defects score were evaluated and the volume of cerebral infarction was observed by TTC staining after 24 h.Nissl staining was used to observe the morphological characteristics of neurons and count the number of surviving neurons.The levels of SOD and MDA were measured.The expression of inducible nitric oxide synthase(iNOS)in cerebral tissue was detected by Western-blot.RESULTS:After treatment of G.biloba extracts,the ethological scoring and volume of cerebral infarction in treatment group were lower than in control group(P0.05).The necrosis and apoptosis of neurons were significantly reduced in G.biloba extracts group.The surviving neurons(66.91±7.53%)in G.biloba extracts group was significantly increased,compared with control group(43.51±4.77%)(P0.05).The serum level of SOD in G.biloba extracts group was higher than in control group while the serum level of MDA was lower than control group(P0.05).The expression of iNOS in control group increased significantly,compared with sham operation group and G.biloba extracts group(P 0.05).CONCLUSION:G.biloba extracts can significantly reduce infarction size,neuronal apoptosis,increase the content of SOD,reduce the content of MDA and inhibit the expression of iNOS.G.biloba extracts have a good neuroprotection effect on rats with focal cerebral ischemia/reperfusion injury in rats.
