湿热型溃疡性结肠炎造模方法的实验研究

目的:复制湿热型溃疡性结肠炎大鼠模型,探讨该模型的客观指标评价体系.方法:采用高脂高糖饮食造成湿热体质,用三硝基苯磺酸(TNBS)和乙醇混合液灌肠诱发湿热型溃疡性结肠炎,观察大鼠证候、结肠病理学改变及血清白介素-6(IL-6).结果:模型大鼠出现粘液便、懒动、食欲减退等湿热证候,病理切片可见结肠粘膜出现明显溃疡、病理学评分有显著性差异(P《0.01),血清IL-6明显升高(P《0.01).结论:采用饮食加化学刺激法首次成功复制出湿热型溃疡性结肠炎的大鼠模型,并拟定证候、病理及免疫三大指标体系,可为临床和实验研究提供参考.

Experimental Study on Animal Model of Damp-heat Ulcerative Colitis

Objective: To make a rat model of damp-heat ulcerative colitis and explore the objective index evaluation of the model.Methods: High fat and sugar were used to induce damp-heat syndrome rats and trinitrotoluene sulfo-acid and ethyl alcohol mixture were used to infuse the test rats to induce damp-heat ulcerative colitis rats.The syndrome rats and the changes of their ulcerative colitis pathology and their blood serum interleukin-6(IL-6) were observed.Results: The model rats showed mucous stool,sluggishness and inappetence.The pathological section showed the obvious ulceration on the colonic mucus.The pathological analysis has a significance difference(P<0.01).The IL-6 was raised obviously(P<0.01).Conclusion: The diet and chemical stimulation method initiatively reduplicates the damp-heat ulcerative colitis rats and makes the index system of syndrome,pathology and immunity,which can provide references for clinic and experiment.
