摘    要:目的建立测定人血清中丙戊酸钠浓度的高效液相色谱(HPLC)。方法将提取的血样用溴苯乙酮在一定条件下衍生化后进行色谱分析。采用Elite C18柱,流动相为甲醇-水(80:20),流速为1mL/min,柱温为室温,检测波长为235nm。结果丙戊酸钠的保留时间为8.14min,最低检出质量浓度为1.64μg/mL,平均回收率为98.37%,其他常用抗癫痫药物不干扰测定,在分析范围内具有良好的线性关系。结论该方法操作简便、快捷、准确,可满足临床快速检测的需要。

Abstract:Objective To establish a method for the determination of sodium valproate in human serum by HPLC. Methods The blood samples were analyzed by chromatography after derivation with bromoacetophenone under certain condition. The Elite C18 column was used with methanol-water(80:20) as the mobile phase and the flow rate of 1 mL/min. The temperature was 10-30 ℃, the wavelength was 235 nm. Results The minimal detectable concentration for sodium valproate was 1. 64 μg/mL. The retaining time of sodium valproate was 8.14 min. The average recovery rate was 98.37%. The other common antiepileptic drugs were not found to interfere with this chromatographic procedure. Conclusion This method is quick, simple, accurate, and suitable for clinical requirements of rapid detection.
