摘    要:目的:探讨吡那地尔预处理对低血容量休克大鼠的心脏保护作用机制。方法:预先给予大鼠吡那地尔(PC组),24小时后复制低血容量性休克模型,在不同时间点内观察吡那地尔对心肌钠泵(Na^+-K^+-ATP酶)、钙泵(Ca^2+-ATP酶)、Mg^2+-ATP酶的活性影响及心肌细胞内游离Ca^2+的水平变化;同时观察正常组和生理盐水对照组大鼠相同时相点的3种酶的活性。结果:PC组心肌离子泵活性均明显高于休克对照组(C组),心肌细胞内游离Ca^2+水平则低于C组;心肌细胞成活率正常组为85%,PC组与C组可达80%。结论:吡那地尔预处理对低血溶量性休克鼠心功能的保护可能与其稳定细胞膜,防止钙超载有关。

Abstract:Objective:To investigate the mechanisms of pinacidil preconditioning on myocardial protection in hypovolemic shock.Methods:Pinacidil was used 24 hours before the establishment of hypovolemic shock model.The activities of Na K ATPase,Ca 2 ATPase,Mg 2 ATPase and free Ca 2 in myocardium were observed at different time points.The activites of ionic pumps were also observed in normal group and in control group at the same time point.Results:Pinacidil preconditioning could increase the activity of ATPase .The concentration of Ca 2 were lower in pinacidil preconditioning group than that in control group.Conclusions:Pinacidil preconditioning can protect myocardial function in hypovolemic shock possibly by stabilizing cell membrane and preventing Ca 2 overload.
