系统性红斑狼疮患者血清中IL-10抑制抗原递呈细胞表面HLA-DR和CD80的表达

目的:研究系统性红斑狼疮(systemiclupuserythematosus,SLE)患者血清中的IL-10对正常单核细胞分化形成的树突状细胞(dendriticcells,DCs)及其前体单核细胞抗原递呈相关膜表面分子表达的影响。方法:分离正常人外周血单个核细胞(PBMC),并采用GM-CSF IL-4方案诱导分化形成DCs,在此过程中加入不同IL-10水平的SLE患者血清或混有IL-10中和抗体的SLE患者血清,联合培养7天后收集细胞,经形态学鉴定,以流式细胞术检测HLA-DR、CD80、CD86的表达。结果:在SLE血清的作用下,DCs的HLA-DR、CD80的表达下调,CD86表达不受影响;中和SLE血清中的IL-10后,HLA-DR、CD80表达能够恢复。其前体细胞单核细胞的HLA-DR、CD80的表达也可被SLE血清下调,用中和抗体中和IL-10后,其表达可部分恢复。结论:高IL-10水平的SLE血清能够抑制DCs及其前体单核细胞表面分子HLA-DR、CD80的表达,对抗原递呈细胞的功能可能产生影响。

Effects of IL-10 in SLE serum on the expressions of HLA-DR and CD80 in monocyte and dendritic cells

Objective:To study the effects of IL-10 in systemic lupus erythematosus(SLE)serum on the expression of antigen-presenting-associated molecula on the membrane surface of monocytes and dendritic cells(DCs).Methods:Normal PBMCs were separated and some of them differentiated into dendritic cells(DCs),induced by GM-CSF and IL-4 in SLE serum including different levels of IL-10.The expressions of HLA-DR,CD80 and CD86 in DCs and monocytes were analysed by flow cytometry.Results:The expressions of HLA-DR and CD80 in DCs were decreased,and the expression of CD86 remained unchanged.After anti-IL-10 antibody was used to neutralize IL-10 in SLE serum,HLA-DR and CD80 expressions were restored.The expressions of HLA-DR and CD80 in monocytes,the precursor cells of DCs,were also down-regulated by SLE serum,but partly restored by neutralization of IL-10 in the serum with anti-IL-10 antibody.Conclusion:The SLE serum with high level of IL-10 can reduce the expressions of HLA-DR and CD80 in DCs and monocytes,which suggests the function of antigen presenting cells might be altered.
