抗人血管内皮生长因子单克隆抗体抑癌作用的形态学观察及作用机制探讨

摘    要:目的 :研究抗人血管内皮生长因子 (VEGF)单抗E1 1 抑制颊癌组织的形态学改变 ,并探讨E1 1 抑癌效应的机制。方法 :接种人颊鳞癌的BALB c裸小鼠分别通过腹腔或瘤周皮下注射抗人VEGF单抗或生理盐水。接种后第 18天处死 ,取颊癌组织进行光镜及透射电镜观察。结果 :E1 1 组颊癌组织中血管分布稀少、存在囊性变坏死区及癌细胞坏死、凋亡等。此外还有血管内皮细胞变性、凋亡及血管壁受破坏、管腔变窄和堵塞等血管系统的异常改变。而生理盐水组癌组织血管丰富 ,颊癌组织结构完整。结论 :抗人VEGF单抗是通过抑制颊癌的血管生成而产生抑癌效应

Abstract:Objective:The purpose of this study was to investigate the mechanism of the inhibited effects of anti_human vascular endothelial growth factor (VEGF) monoclonal antibodies E\-\{11\} on the growth of buccal mucosal carcinoma. Methods:E\-\{11\} was hypodermic and celiac injected into BALB/c nu/nu mice, which were transplanted with buccal carcinoma. The saline was injected as the negative control. Mice were killed on the 18th day and the pathological alteration of tumor was evaluated. Results:Microscopically, massive necrosis and decreased vascular density was observed in tumor tissues treated with E\-\{11\}. Correspondent with the aforementioned phenomena, degeneration and apoptosis was also found in the tumor cells. For the first time, degeneration and destruction of the vascular endothelial cells in the tumor tissues treated with E\-\{11\} were observed using microscopy and transmission electron microscopy examination. Conclusion:The study demonstrated that E\-\{11\} inhibited the tumor angiogenesis by blocking the action of VEGF and resulted in significant inhibitory effects on the tumor growth. The result would offer a therapeutic base to the clinical application of the anti_human VEGF monoclonal antibodies.
