垂体腺瘤中Bcl-2和Bax的表达与其侵袭性的关系

摘    要:目的通过对垂体腺瘤中Bcl-2和BaxmRNA和蛋白表达水平的检测,探讨它们与垂体腺瘤侵袭性之间的关系。方法RT-PCR检测8例垂体腺瘤中Bcl-2和BaxmRNA的表达,免疫组化检测78例垂体腺瘤中Bcl-2和Bax蛋白的表达,分析侵袭组和非侵袭组垂体腺瘤之间Bcl-2和BaxmRNA和蛋白表达的差异和相关性。结果侵袭组中Bcl-2的表达较非侵袭组显著增高,而Bax的表达差异无显著性。Spearman等级相关分析显示,Bcl-2蛋白表达的阳性率与Bax蛋白表达呈负相关关系。结论Bcl-2在侵袭性垂体腺瘤中表达水平显著增高,而Bax的表达差异无显著性,提示垂体腺瘤侵袭性生长主要是由于Bcl-2的抗凋亡效应的增强,并非Bax的促凋亡效应减弱的结果。

Abstract:[Objective] To explore the relationship between Bcl-2 and Bax expressions with invasiveness of pituitary adenomas. [Methods] The mRNA expressions of Bcl-2 and Bax were measured by RT-PCR in 8 pituitary adenoma samples. Immunohistochemical study for protein expressions of Bcl-2 and Bax in total 78 patients were divided into invasive and non-invasive pituitary adenoma groups. [Results] The Bcl-2 mRNA and protein expressions in invasive group were significant higher than those in non-invasive group, however, the Bax mRNA and protein expressions did not significantly differ between the two groups. The Bcl-2 and Bax protein expressions in pituitary adenomas were inverse correlated. [Conclusion] These data implicates that Bcl-2 inhibit cell apoptosis relatively increases the proliferative activity of pituitary tumor and results in invasive growth. At the same time, it showed that the invasive behavior of pituitary adenomas results mainly from the increased anti-apoptotic activity of Bcl-2, but not the decreased pro-apoptotic activity of Bax.
