附子干姜不同药对配伍提取物对小鼠脾细胞增殖的影响

摘    要:目的:观察四逆汤组方附子和干姜的不同药对配伍提取物对小鼠脾细胞增殖的影响。方法:附子(F)分别与葱白(B)、甘草(C)、猪胆汁(D)、干姜(G)、人参(R)配伍和G分别与B、C、D、R配伍的乙醚、氯仿、水提取物为研究对象,采用小鼠脾细胞为实验模型,MTT法测定细胞内线粒体脱氢酶活性。结果:与对照组比较,乙醚提取物中与附子配伍的FB、FC、FD、FG、FR组均有刺激脾细胞增殖作用(P<0.05,P<0.01),与干姜配伍中只有GR组有刺激脾细胞增殖作用(P<0.05)。氯仿提取物中FG、FR组有刺激脾细胞增殖作用(P<0.05,P<0.01)且刺激作用强于F组(P<0.05)。水提取物中与附子配伍的各组均有刺激脾细胞增殖作用(P<0.05,P<0.01),与干姜配伍各组刺激作用较弱(P<0.05),且FB、FC、FG、FR组刺激作用强于F组(P<0.05,P<0.01)。结论:与附子配伍的药对提取物活性较强,与干姜配伍活性较弱,体现出附子配伍的君药作用。与附子配伍的氯仿、水提取物活性强于附子,揭示药对配伍增效作用。

Abstract:Objective:To study the effects of extractions from different drug pairs of aconite root(F)and dried ginger(G)on proliferation of mice spleen cells in vitro.Methods:The cultured mice spleen cells were exposed respectively to ether or chloroform or water extractions from different drug pairs of F matching respectively with fistular onion stalk(B)or liquorice root(C)or pig bile(D)or dried ginger(G)or ginseng(R)and drug pairs of G matching respectively with B or C or D or R for 72h,and then the activities of intracellular mitochondria dehydrogenase were investigated by methyl tetrazolium assay.Results:Compared to the control group,ether extractions from FB,FC,FD,FG,FR and chloroform extractions from FG,FR and water extractions from FB,FC,FD,FG,FR could increase dehydrogenase activity significantly(P<0.05,P<0.01).Ether extractions from GR and water extractions from GB,GC,GD,GR could significantly increase dehydrogenase activity(P<0.05).Compared to the F group,chloroform extractions from FG,FR and water extractions from FB,FC,FG,FR could increase dehydrogenase activity more significantly(P<0.05,P<0.01).Conclusion:The activities of extractions from drug pairs compatible with aconite root are stronger and the activities of extractions from drug pairs compatible with dried ginger are weaker,which shows the monarch drug role of aconite root in compatibility with other drugs.The activity of chloroform extractions and water extractions from drug pairs compatible with aconite root are respectively stronger than that of chloroform extractions and water extractions from aconite root,which suggests the synergism effect of drug pairs.
