茵陈对肝内胆汁淤积湿热证大鼠肝组织NTCP表达的影响

摘    要:目的:通过研究茵陈治疗后,肝内胆汁淤积湿热证大鼠肝组织中钠-牛黄胆酸盐共转运多肽(Na+/tauro-cholate Cotransporting Polypeptide,NTCP)的表达,从而探讨茵陈的利胆退黄的作用机制。方法:选用雄性SD大鼠45只,随机分为3组:正常组、模型对照组和茵陈组。观察大鼠的体重、肝功能、肝组织病理变化和肝脏NTCP的表达。结果:治疗后茵陈组大鼠的体重(199.11±3.54)g高于模型对照组(182.80±5.89)g,P<0.05。茵陈组大鼠血清TB、DB、ALT、AST和ALP水平分别为(17.76±3.72)μmol/L,(10.17±1.47)μmol/L,(77.18±6.86)U/L,(218.18±32.06)U/L,(230.49±28.84)U/L],明显低于模型对照组(34.72±3.65)μmol/L,(19.33±3.27)μmol/L,(123.73±18.51)U/L,(353.78±63.08)U/L,(355.49±63.08)U/L],P<0.05。苏木精-伊红染色法显示茵陈组大鼠肝细胞的坏死、炎症、水肿程度明显好于模型对照组大鼠。免疫组化染色示茵陈组大鼠NTCP阳性表达肝细胞表达多于模型对照组。荧光定量RT-PCR检测显示茵陈组大鼠肝细胞NCTP mRNA表达(2-△△CT值为2.46±0.71)高于模型对照组(2-△△CT值为1.48±0.38),P<0.05。结论:茵陈能增加肝内胆汁淤积湿热证大鼠的体重,有效改善胆汁淤积、肝功能及减轻肝脏病理损害。茵陈能提高肝内胆汁淤积湿热证大鼠肝细胞NTCP的表达。

Abstract:Objective:To investigate the choleretic and jaundice-relieving mechanism of Yinchen and the NTCP expression levels in liver of rats with intrahepatic cholestasis and damp-heat were detected after treatment with Yinchen.Methods:Forty-five male SD rats were randomly assigned into 3 groups:normal,model control and Yinchen groups.The body weights,liver funtions,liver pathologies and NTCP expressions in rat livers were detected.Results:The rat body weights in Yinchen group (199.11±3.54)g] were greater than model control group after treatment (182.80±5.89)g],P<0.05.The blood serum levels of TB,DB,ALT,AST and ALP in Yinchen group(17.76±3.72μmol/L,10.17±1.47μmol/L,77.18±6.86U/L,218.18±32.06U/L,230.49±28.84U/L)were lower than these in model control group(34.72±3.65μmol/L,19.33±3.27μmol/L,123.73±18.51U/L,353.78±63.08U/L,355.49± 63.08U/L),P<0.05.The degrees of necrosis,inflammation and edema of rat hepatocytes in Yinchen group were mild than these in model control group as shown in the hematoxylin-eosin staining.More NTCP postive hepatocytes were found in rats of Yinchen group than these of model control group as shown in the immunohistochemical staining.In the analysis of fluorescent quantitative RT-PCR,the NTCP mRNA expression levels of rat hepatocytes in Yinchen group(the value of 2-△△CT was 2.46±0.71)were higher than these of model control group(the value of 2-△△CT was 1.48± 0.38),P<0.05.Conclusion:Yinchen can increase the body weight,improve cholestasis and liver function and mitigate pathological damage of rat with intrahepatic cholestasis and damp-heat.Yinchen can enhance the NTCP expression levels in the hepatocytes of rat with intrahepatic cholestasis and damp-heat.
