清肠败毒饮对溃疡性结肠炎大鼠肠黏膜组织NF-κB水平的影响

摘    要:
目的:观察中药清肠败毒饮对实验性溃疡性结肠炎(UC)大鼠的治疗作用,并探讨其可能的作用机制。方法:用三硝基苯磺酸(TNBs)法复制大鼠实验性溃疡性结肠炎模型,分别给予清肠败毒饮和柳氮磺吡啶(SASP)治疗,14d后处死.观察结肠黏膜形态学和病理组织学变化,用免疫组化法测定大鼠结肠黏膜组织NF-κB的表达情况。结果:清肠败毒饮可显著改善实验性UC大鼠结肠黏膜病变,降低NF-κB的表达,与模型组比较差异有统计学意义(P〈0.05)。结论:清肠败毒饮能减轻实验性UC大鼠结肠黏膜损伤,促进炎症吸收和组织修复,推测其作用机制可能是与降低NF-κB表达水平有关。

Abstract:
Objective:To observe the therapeutic effect and mechanism of Qingchang Baidu Decoction on rats with ulcerative colitis (UC).Methods :40 rats were randomly divided into normal control group,model group,Qingchang Baidu decoction group and salicylazosulfapyridine groul.The ulcerative colitis rat models were induced by 2,4,6-trinitrobenzenesulfnic acid (TNBs).The morphological and histological changes of colonic mucosa of rats in 4 groups were observed under microscope, the expression of NF-κB in colonic mucosa was detected by immunohistochemistry.Results:Compared with the model group,the expression of NF-κB in colonic mucosa was markedly decreased,the pathological changes of colonic mucous were significantly relieved in Qingchang Baidu decoction group.Conclusions:Qingchang Baidu Decoction could alleviate the injury of colonic mucosa,accelerate the absorption of inflammation and the rehabilitation of tissue.Its mechanism may be related to the decrease of the expression of NF-κB.
